Ganoderma polysaccharides, GLB, are a fraction of polysaccharides isolated from the fruiting body of Ganoderma lucidum (Leyss ex Fr. Karst.), which are water-soluble. Numerous studies have demonstrated that Ganoderma polysaccharides have immunomodulatory and anti-tumor effects and the majority of reports implied that Ganoderma polysaccharides have no direct inhibitory or killing effects on tumor cells, whereas the polysaccharides show marked anti-tumor effect in vivo. In this regard, it is generally believed that the anti-tumor effect of Ganoderma polysaccharides is mediated through enhancement of the immune function. A lot of researchers have studied the mechanism of anti-tumor action of Ganoderma polysaccharides from immunological perspective and made remarkable progress, but the exact mechanism is still unclear. It is, therefore, necessary for the mechanism to be studied further.OBJECTIVE: It was attempted to explore the immunological mechanism of anti-tumor action of Ganoderma polysaccharides from the aspects of Toll-like receptor 4 (TLR4) and its signal transduction, and the possibility of antagonizing the inhibitory effect of prostaglandin E2 (PGE2) on immune function by using tumor-bearing mice and their immune cells for the experiments.METHODS: MTT colorimetry was used for the assay of cell proliferation.
