Objective To investigate the expression of pituitary tumor-transforming gene(PTTG-1)and basic fibroblast growth factor(bFGF)in uterine leiomyoma cells and the relationship of them.Methods Uterine leiomyoma cells and normal myometrial cells were subjected to primary culture and were verified by immunocytochemically(SABC).Semi-quantitative RT-PCR was used to detected expression of PTTG-1 mRNA and bFGF mRNA in these two cell lines and analysised the expression of PTTG-1 mRNA after bFGF treatment.Result(1)The cells from uterine tissue specimens could be cultured successfully and verified by immunocytochemically.(2)The expression of bFGF mRNA is higher in leiomyoma cells than in normal myometrial cells.There were statistic difference between them(P<0.05).(3)PTTG-1 mRNA level in leiomyoma cells was significantly higher than in normal myometrial cells(P<0.001).The levels of PTTG-1 in the secretory phase and in the proliferative phase is no significant difference(P>0.05).(4)Leiomyoma cells were treated with bFGF for various concentration(0,5,10,50ng/mL).There was significant difference in the four groups by analysis of variance,the levels of PTTG-1 has a significantly increased with the increasing of concentration(P<0.01).(5)bFGF(Sng/mL)induced PTTG-1 expression.At 24h,36h after bFGF treatment,the expression of PTTG-1 was elevated,but did not reach statistical significance (P>0.05).At 48h,60h,its expression was significantly increased.There were statistic difference (P<0.05).Conclusion(1)bFGF as an important growth factor can improve the development of leiomyomathrough fastening vascular formation and stimulating growth of leiomyometrial cells.(2)The expression of PTTG-1 is significantly elevated in leiomyoma and related closely to the development of leiomyoma.It is independent of the menstrual cycle and is not significantly affected by ovarian hormones.(3)bFGF time- and dose-dependently stimulates PTTG-1 expression.The positive feedback loop between them may be pivotal in the growth of leiomyoma cells.
