Objective To study the correlation between kidney deficiency syndrome and apolipoprotein E (ApoE) gene polymorphism in postmenopausal women.Methods According to Tranditional Chinese Medicine colleges and universities teaching reference book-《Diagnostics of Chinese Medicine》and《National standards of Syndrome Clinic》, the Kidney Syndrome questionnaire were preparate. We carried on the Kidney Syndrome questionnaire survey to permanent women residents of Nanning City that they Carries on physical examination in the Chinese medicine out-patient service and cadres physical examination of our hospital in May 2008 to December 2008 and their ages=50 years and menopause for 5-20 years. According to《Tranditional Chinese Medicine deficiency dialectical reference standard》We randomly selected 80 women patients with kidney deficiency syndrome as a kidney observation group,65 cases of have not kidney deficiency syndrome of women as a normal control group. Meanwhile, Patients were excluded who have serious disease of circulation,liver,spleen,lung,kidney and endocrine system. PCR-restriction fragment length polymorphism (PCR-RFLP) was used to detect ApoE genotype and allele frequencies of 80 postmenopausal patients with kidney deficiency syndrome and 65 patients without kidney deficiency syndrome.Statistical Methods:used SPSS 16.0 software to data process.Classification statistic was conducted on kidney syndrome and symptoms.Gene frequencies was counted by gene counting method and subjects with the Hardy-Weinberg equilibrium in line with the x2 test level.Compare with ApoE genotype and allele frequencies between observation group and control group By the x2 test. Comparison with ApoE genotype and allele frequencies between different symptoms of observation group used the x2 test。Results The test results and statistical analysis of ApoE as follows: (1)We can detect five genotype in observation group:ε2/ε3,ε2/ε4,ε3/ε3,ε3/ε4,ε4/ε4,frequency of them was 11.25%,8.75%,58.75%, 11.25%,10.00%;while four genotype in control group were deceted:ε2/ε3,ε2/ε4,ε3/ε3,ε3/ε4,frequency of them was 15.38%6.15%,69.23%,9.24%.in observation group,ε3/ε3 genotyoes is the most,followed byε2/ε3 genotypes;in control group,ε3/ε3 genotypes is also the most.,followed byε2/ε3 genotype,andε2/ε2 genotypes is rare.We couldn't detectε2/ε2 genotypes in observation group and control group.Frequency of ApoE alleleε2,ε3 andε4 in observation group was 10.00%,70.00%and 20.00%,while control group was 10.76%, 81.55%,7.69%.(2)Frequency ofε4/ε4 genotype in observation group has significantly different with control group(P<0.05),frequency of ApoE alleleε4 in observation group has significantly different with control group(P<0.05).(3)Incidence of relative risk (OR value)of ApoE alleleε4 and kidney deficiency syndrome is 3.33>1.Conclusion(1)ApoE Alleleε4/ε4 genotype is related to kidney deficiency syndrome in postmenopausal women, and ApoE Alleleε4 is closely related to kidney deficiency syndrome in postmenopausal women. Increased frequency of ApoE Alleleε4 may be a risk factor for kidney deficiency syndrome. (2)ApoE genotypeε4/ε4 is a useful marker for detection and aided diagnosis in kidney deficiency syndrome of postmenopausal women. (3)There has a significant correlation between the occurrence of kidney deficiency syndrome andε4 allele contained. From that,we can confirm objective clinical measurement value of kidney deficiency syndrome diagnosis from genetics and gene,it can provide a scientific basis for objective diagnosis of kidney deficiency syndrome and research approach and basis on intervention to risk factors of kidney deficiency syndrome and treatment based on syndrome differentiation of kidney deficiency syndrome.
