The Effects Of Pretreatment Acupoint Injection Of Xiangdan Parenteral Solution At Neiguan (PC6) On Fibrinolytic System T-PA,PAI-1 In Acute Myocardial Ischemia Rats

Objective:Through injecting HongDan parenteral solution into Neiguan point of acute myocardial ischemia rats to understand how HongDan parenteral solution working on t-PA,PAI-1 activity of endothelium-dependent's fibrinolytic system. And to discuss the influence on fibrinolytic activity, as well as the prevented and therapeutic effects on ischemia myocardial of acupuncture combined with medicine group. What's more, it may help providing some new ideas and methods for clinical treatment of the coronary disease and ischemia myocardial.Methods:SD rats were pretreated for 14 days by injection of HongDan parenteral solution into Neiguan point, before copying ischemia myocardial model. Then the ischemia myocardial model was copyed by peritoneal subcutaneous injecting isoproterenol(ISO).The myocardium pathomorphology was observed, the t-PA, PAI-1 protein of coronary artery blood vessel endothelium were detected by immunohistochemisty. The blood plasma t-PA, PAI-1 of fibrinolytic system were detected by the ELISA of enzyme unite.Results:(1) Ischemia and hypoxia of myocardium were observed in the acute myocardial ischemia rats. The acupuncture combined with medicine group can significantly reduce the cardiac damage to the acute myocardial ischemia rats. It also can decrease the size of myocardial ischemia, and alleviate the degree of myocardial ischemia, to speed up restoration. The medicine group, acupoint injection group and control group didn't significantly improve the myocardial damage. And the above-mentioned effect of acupuncture combined with medicine group is much better than the Acupoint injection group and the medicine group. (2) The expression of t-PA was lower after ischemia mocardial, and the acupuncture combined with medicine group, acupoint injection group and the medicine group can improved it, especially the acupuncture combined with medicine group.(3)The expression of PAI-1 was increased after ischemia myocardial. And the acupuncture combined with medicine group, acupoint injection group and the medicine group can lower the expression, whereas the ampoul group is the best. (4) Compared with model group, significant differences were observed in the acupuncture combined with medicine group, acupoint injection group and the medicine group regarding t-PA/PAI-1. Significant differences between acupuncture combined with medicine group and acupoint injection group and the medicine group were observed regarding t-PA/PAI-1. There is no difference between acupoint injection group and medicine group.Conclusions:Pretreatment of injecting HongDan parenteral solution into Neiguan point can prevent the cardiac muscle of acute myocardial ischemia rats. The pretreatment can raise the contents of t-PA and decrease the contents of PAI-1, systemicly and locally in coronary artery. It also can lighten myocardial injury in acute myocardial ischemia. Furthermore, the acupoint injection can be regarded as a pretreatment for acute myocardial ischemia, increasing the survivability of animals' to ischimia myocardial, improving the activity fibrinolytis systemicly and locally, to lighten the damage after the ischemia of the myocardial.
