Objective:A common feature of athletics is full of drastic conflictions and competitions.The ankle parenchyma of athletes may get hurt in certain sports because of one over-extent action or a long time intensive training,which is easy to cause arthropathy and will not heal in a short time. In some serious condition,it even causes deformity.Joint inflammation is the most important reason for joint injury. Cytosolic Ca2+ overload has been proposed as one of the prime factors for many inflammatory cells activation and the relative enzymes and inflammatory factors release.It is therefore accepted that Ca2+ channel blockers can protect joint from inflammation.In the present studies,we investigated the possible molecular mechanism of inflammatory arthropathy,and the antinociceptive efficacy of HWTX-Ⅰ,a spider peptide toxin blocking Ca2+ channels,on rat models with adjuvant arthritis.Methods:64 grown-up male SD rats,which were randomly divided into the sham-operation group,the physiological saline group, the HWTX-Ⅰgroup and ibuprofen group.We induced rat models of adjuvant arthritis combined with Yaksh T.L,Rudy T.A catheterization in spinal epidural space in the study,relative anti-inflammatory drugs were given to rats through the reserved PEIO plastic tubes or intragastric administration.RT-PCR was used to detect the level of TNF-α,IL-1β,IL-6 expression in synovium and chondrocyte of inflammatory joints,and ELISA was used to detect the concentration of TNF-α,IL-4,IL-10 in serum Results:(1) The result of pain test of joints injured side shown that, HWTX-Ⅰcan obvious relieve joint analgesia after epidurally administered,and was more effective than ibuprofen.(2) The result of circumference measurement of joints injured side shown that,HWTX-Ⅰhad obvious anti-arthredema efficacy after epidurally administered,and was more effective than ibuprofen.(3) The results of ELISA shown that TNF-αlevel of physiological saline group showed an obvious increase compared with HWTX-Ⅰgroup and ibuprofen group(P<0.05).However,the level of IL-4 in physiological saline group was significant difference from HWTX-Ⅰgroup(P<0.05) and ibuprofen group(P<0.01).The IL-10 lever in physiological saline group also showed significant lower than ibuprofen group(P<0.05) and was very significantly different from HWTX-Ⅰgroup(P<0.01), IL-10 lever in HWTX-Ⅰgroup was the highest and was significant difference from ibuprofen group(P<0.05) too.(4) RT-PCR have shown that TNF-α,IL-1β,IL-6 mRNA expressed in physiological Saline group are much more higher than ibuprofen group and HWTX-Ⅰgroup(P<0.01),we can also find the inhibition of HWTX-Ⅰis more effective than ibuprofen.Conclusion:(1) In our research,we evaluated the ethology of rats with pain test,the result showed that HWTX-Ⅰhad obvious analgesia efficacy compared with physiological saline.(2) The ankle joint circumferences have been measured,the result showed that HWTX-Ⅰcould eliminate arthrocele in some degree.(3) The administration of HWTX-Ⅰin spinal dura mater space can down regulation the concentration of TNF-αin rats serum, and up regulation the concentration of IL-4,IL-10 in serum. HWTX-Ⅰis more effective compared with ibuprofen,which is common used as calcium channel blocker in clinic.The research proved that HWTX-Ⅰhas obvious antinociceptive efficacy.(4) The administration of HWTX-Ⅰin spinal dura mater space can down regulation the expression of mRNA of inflammatory cytokines(TNF-α,IL-1β,IL-6) in inflammatory joints.HWTX-Ⅰis more effective compared with ibuprofen,which is common used as calcium channel blocker in clinic.The research proved that HWTX-Ⅰcan relieve and inhibit the pathogenesis of inflammatory arthropathy.(5) The movement of calcium has high correlated with the development of inflammation.Our research has approached the molecular mechanism of inflammatory arthropathy and the efficacy of calcium channel blocker,it is significance for prevention and cure of joints inflammation caused by sports.
