Effect Of Exercise Intervention On Inflammatory Cytokines In Negative Emotion Stimulated Aged SD Rats

Research purposes: The purpose of this study is to stimulate aged rats by long-term negative emotion, so as to establish the inflammatory immune suppression model resulting from negative emotion stimulation,then moderate-intensity aerobic swimming intervene inflammatory immune suppressive aged rats to observe the negative emotion stimulation and exercise intervention on the impact of inflammatory cytokines.Research Methods: 48 Healthy aged male SD rats, around the age of 15 months, 1 week after adaptation, in which 24 rats were randomly selected as negative emotional stimulation group, and the other 24 rats as the control group. After 3 weeks, in which 24 negative emotional stimulation rats were randomly selected 8 as negative emotional stimulation group(P 1), in which 24 control group rats were randomly selected 8 as three weeks quiet control group (C1) for analysis of inflammatory factors. After Model is successful, the remaining rats were divided into four groups: 9 weeks Quiet control group (C2), 3 weeks quiet rest and 6 weeks negative emotional stress and group (P2), 3 weeks quiet rest and 6 weeks exercise n group (E), three weeks negative emotion stimulation and 6 weeks exercise intervention group (P2E). IgG, IgA, IgM, CRP measured by turbidimetry ;Cor, IL-1β, IL-6, TNF-αmeasured by RIA.Results:(1) Compared to C1 group, P1 group Cor, IL-6, CRP increased very significantly(P<0.01);TNF-α,IL-1β,IgA significantly increased(P<0.05);IgG increased slightly, IgM decreased slightly but not significantly. these prompted 3 weeks negative emotion stimulation made the increase of inflammatory factors.(2) Compared to C2 group, P2 group Cor and CRP are significantly increased(P<0.01);IL-1βincreased significantly(P<0.05);IgA decreased significantly(P<0.05);IgG and IgM decreased slightly but not significantly. These prompted passive rest after the negative emotion stimulation can not make high level state of the inflammatory cytokines restore to the level of the control group.Antibody levels can not return to the level of the control group.(3) Compared to C2 group,E group Cor and TNF-αare increased very significantly (P <0.01); CRP increased significantly (P <0.05);IgG, IgA, Ig M, IL-1β, IL-6 did not change significantly. These prompted aerobic exercise led to change in stress hormone, TNF-α, CRP increased, other inflammatory factors did not change significantly.(4) Compared to C2 group, P2E group Cor and CRP are increased very significantly (P <0.01); IgG was increased significantly (P <0.05), IgA, Ig M, IL-1β; IL-6, TNF-αdid not significantly change. Exercise intervention reduced that negative emotion can stimulate the inflammatory role, made content of inflammatory factors close to the level of a control group.(5) Compared to P2 group, P2E group Cor ,IgG and IgA are increased very significantly (P <0.01), IL-1β, IL-6 and Ig M decreased slightly but not significantly, TNF-αincreased slightly but not significantly. These prompted aerobic exercise is more effective than passive rest to slow down inflammatory response of the negative emotion in old rats.(6) Compared to E group, P2E group Cor and CRP are increased very significantly (P <0.01), TNF-αwas significantly decreased (P <0.01), IL-1βand IL-6 decreased slightly but not significantly, IgG significantly increased (P <0.05), IgA and Ig M increased slightly but not significantly.these note that exercise have significant role on old rats with negative emotion stimulation.Conclusion: (1)The negative emotion stimulation of three weeks made inflammation markers increased, the negative emotion stimulation that can effectively establish the model of inflammatory immune suppression in rats.(2) After the negative emotion stimulation, passive rest can not slow down inflammatory response. Active intervention should be to find ,which as a means to offset the negative effect of psychological stress and prevent accelerated aging.(3) Aerobic exercise can reduce the inflammatory response. Exercise can interfere with the high level status of pro-inflammatory cytokines which was caused by negative emotion stimulation and increase antibody levels.(4) Aerobic exercise is more effective than quiet rest to slow down the inflammatory response of the aged rats which was caused by negative emotion stimulation.(5) Exercise intervention with negative emotion stimulation than the aerobic exercise without negative emotion stimulation more effectively reduced the level of the inflammatory response in old rats.The effect of exercise intervention is more significant, the reason is that the emotional stimulation increase the level of inflammatory makers.
