Background: Cisplatin is a platinum complex that binds to DNA to form cross-bonds that inhibit DNA replication.High doses of Cisplatin inhibit RNA and protein synthesis.Cisplatin is a periodic non-specific anti-tumor drug.It has a broad anticancer spectrum and is effective against a variety of solid tumors,such as testicular tumor,breast cancer,lung cancer,head and neck cancer,ovarian cancer,osteosarcoma and melanoma.It is currently one of the most commonly used drugs in combination chemotherapy.However,like other chemotherapy drugs,cisplatin has a number of toxic side effects,including toxicity to the kidney,nervous system and pancreas.These toxicity will not only interfere with the progress of treatment,but also greatly reduce the quality of life of patients.Therefore,how to reduce the toxic and side effects of chemotherapeutic drugs and improve the quality of life of patients is a research topic that attracts much attention at present.In recent years,many scholars have found that cisplatin can cause severe inflammatory reactions in the body.Various cytokines are produced during the inflammatory response.In the physiological state,cytokines play an important role in maintaining the self-stability and function of the body's immune system,but in the process of chemotherapy,the inflammatory response induced by cisplatin chemotherapy drugs may be related to its toxic and side effects.Aerobic exercise is a widely used non-drug intervention at the present stage and has been increasingly applied to clinical intervention and rehabilitation treatment.A large number of studies have shown that exercise can improve the related side effects caused by cisplatin chemotherapy.The mechanism of action may be to improve the inflammatory response by regulating the level of inflammatory cytokines in the serum to achieve the effect,but more research is needed on the specific mechanism of action.Objective: To investigate the effect of moderate-intensity aerobic exercise intervention on the quality of life and the level of inflammatory factors induced by cisplatin in rats,so as to accumulate more experimental data for future research.Methods:In 4 to 5 weeks of SPF male rats(30)as the research object,30 rats wererandomly divided into 3 groups: control group 10(N),cisplatin chemotherapy group(A)only10,sport + cisplatin chemotherapy group(B)only 10,A,B two groups weekly cisplatin intraperitoneal injection,intraperitoneal injection of A 4 mg/kg,group N physiological saline injection,A total of around;After each cisplatin injection,group B began to perform moderate intensity aerobic exercise intervention on the following day.The exercise mode was platform training,with platform slope of 0° and initial speed of 15 m/ min,followed by an increase of 1m/ min until 19 m/ min,each speed of 10-15 min,5 times a week for 4 weeks.During the experiment,the living conditions of the rats were closely observed,the intake and weight of the rats were tested weekly,and the strain test was conducted.After the experiment,the serum and bilateral gastrocnemius muscles of the rats were sacrificed to detect the wet weight ratio of the gastrocnemius muscles of each group.Serum levels of Il-6?Il-1??Il-10 and TNF-?were detected by ELISA.Research results:(1)Compared with group N,food intake and body weight in group A decreased significantly after chemotherapy,and rats in group B increased food intake and body weight after 4 weeks of moderate intensity aerobic exercise intervention.(2)Compared with group N,the tension in group A decreased significantly after chemotherapy,while that in group B increased after 4 weeks of moderate intensity aerobic exercise intervention.(3)Compared with group N,after chemotherapy,the wet weight ratio of gastrocnemius muscle in group A was significantly lower than that in group N(P < 0.05).After 4 weeks of moderate intensity aerobic exercise intervention,the wet weight ratio of gastrocnemius muscle in group B was increased(P < 0.05).(4)Compared with group N,serum levels of Il-6 in group A and group B increased after chemotherapy(P < 0.05).After 4 weeks of moderate intensity aerobic exercise intervention,serum levels of Il-6 in group B decreased compared with group A(P < 0.05).Compared with group N,serum Il-1? levels in group A and group B increased after chemotherapy,and there was A significant difference between group A and group N(P < 0.01).After 4 weeks of moderate intensity aerobic exercise intervention,serum Il-1? levels in group B decreased compared with group A(P < 0.05).(5)Compared with group N,serum Il-10 levels in group A and group B increased after chemotherapy(P < 0.05),and there was A significant difference between group A and group N(P < 0.01).After 4 weeks of moderate intensity aerobic exercise intervention,serum Il-10 levels in group B decreased compared with group A,with no difference(P > 0.05).Compared with group N,serum levels of TNF-?in group A and group B increased after chemotherapy(P < 0.05).After 4 weeks of moderate aerobic exercise intervention,serum levels of TNF-?in group B decreased compared with group A(P < 0.05).Research conclusions:(1)Cisplatin caused reduced food intake,weight loss,muscle mass and muscle strength in normal rats,and the expression levels of serum pro-inflammatory cytokines Il-1??TNF-??Il-6 and Il-10 were up-regulated in rats.(2)After the intervention of moderate intensity aerobic exercise,the serum levels of Il-1??TNF-??Il-6 and other inflammatory factors decreased,suggesting that the intervention of moderate intensity aerobic exercise may be a mechanism to reduce the inflammatory response and improve the quality of life.
