Paclitaxel (taxol) is an important natural anti-cancer product with efficient clinical effect which extracted from the rare plant yew. It has been permitted to treat ovarian and breast cancer by many countries. At present, it is largely extracted from yew. For the low content of the Paclitaxel and existence of some analogues of the Paclitaxel in the yew, it is very difficult to separate it. Consequently, it is very important to develop an effective method of separation and purification Paclitaxel, to reasonably use the limited resources.From the condition of recyclation of plant resouces and industrial production, this experiment carried out a detail research of direct extraction of Paclitaxel from yew. Meanwhile, the method of extracting 10-DAB procursor of semisynthesis of paclitaxel was investigated. Because the total recovery of both Paclitaxel and 10-DAB are much more 65% and the final purity of them are higher 98%, both of them can be used as raw materials of medicine. The method was proved efficiently to extract Paclitaxel from yew.Three experiment stages were designed in the study. Firstly, using processed yew branches and leaves as raw material to extract rude Paclitaxel by seepage flow in room temperature;Secondly, by solid phase extraction, liquid-liquid extraction, chromatography, bromination, second chromatography, cystallization and finally second cystallization to purifythe rude Paclitaxel to a purity reaching 99.13%;lastly, the water phase , which from the first liquid-liquid extraction, as raw material, was processed throught liquid-liquid extraction, saponification, then chromatography, re-chromatography, cystallization and re-cystallization to get 10-DAB of purity 99.02%.By TLC and HPLC on-line detection the data from the experiment show precise and accuracy. It also gives full consideration of the practical feasibility from pilot to industrial production in the research.
