| In the study, 6 pairs microsatellite primers(S0155, SW240, IGF1, SW951, SW857, SW24) were selected in accordance to recommendation of Food and Agriculture Organization of the United Nations (FAO) and 27 pairs microsatellite primers of swine from literature. Microsatellite DNA detection was applied to 3 populations(90 samples) Guizhou local swine expecily Nuogu Pig with the filtered primers to research genetic structure and genetic relationship of Guizhou local swine in molecular level. The results showed that:1. At 6 microsatellite loci in 3 populations of Guizhou local swine; 43 alleles were found of which the shared were 16 and the unshared were 24. Allele frequency was between 0.0089-0.5241. The alleles which had high frequency possessed strong genetic superiority and genetic stability, embodied genetic traits of Guizhou local swine.2. The percentage of all polymorphic loci of 6 pairs primers in 3 population reached 100%. Observed heterozygosity, expected heterozygosity, Nei's expected heterozygosity were 0.9278±0.2109, 0.7987±0.0294,0.7975±0.0287, 0.7605±0.0318 respectively.3. Mean effective allele were 5.3±0.9513, and mean polymorphism information content(PIC) were 0.7196±0.0417 and high polymorphism which accounted for abundance in genetic diversity of Guizhou local swine; Both GD and SI of Nuogu Pig are highest (GD:0707 ,SI:1.431),three types of loci swine GD(0.611-0.689 ),SI(1.112-1.342).4.Genetic variance from high to low is Nuogu Pig ,Kele Pig,Guanling Pig.This is derived from MNA, Ne, PIC , GD and SI; The Nei standard genetic distance of three swine is 0.0965 -0.3080.The fartherst distance is Nuogu Pig and Guarding Pig, the nearest distance is Nuogu Pig and Kele Pig.The distance is 0.7346-0.9050.5.On the basis of abundance in genetic diversity of Guizhou local swine, high genetic heterozygosity, and actuality of Guizhou swine resources, proposing that conservation of Guizhou local swine should be combined with development and employment organically, so that development and employment facilitated conservation; Nayong "Nuogu Pig" is a specile type of Kele Pig .The study supplied further genetic evidence for conservation and development of Guizhou local swine. |
