Different Origin Of Pinellia Germplasm Preliminary Evaluation Studies

Pinellia tuber,the tuber of Pinellia ternata(Thunb.) breit.,is one of blindly used bulk herbs.Although the natural resources of Pinellia ternata is a very Wider distribution,it sharp decline year by year.The wild and cultivation resources hardly meet market demand.The pros and cons of the quality of germplasm is important for Chinese herbal medicines,and have an significance on breeding.Genetic Diversity of germplasm resources is not only the diversity of a high-quality ingredients,but also is fundamental to the protection of resources and the introduction of Chinese herbal medicines on the premise.The work in this thesis is part of the National 11th Five-Year scientific and technological support projects:Pinellia tuber genuine medicine-producing standardized production research of key technologies.Through the investigation of national resources and production status of Pinellia ternata in China,carried out the wild and cultivated resources for the collection,comparison of different origin of the differences in germplasm,breeding work to lay the foundation.It's important for germplasm resources for the protection of traditional Chinese medicine,improve the ecological environment,sustainable use of resources, and ensure the safety of drugs and Pinellia ternate industrial development is of great significance.We study on germplasm evaluation,Firstly,collection of 8 provinces in 17 places of 17 germptasm,Then,records of the investigation in the natural climatic conditions and ecological environment collection sites Pinellia ternate, all localities Pinellia ternate germplasm sources,the local Pinellia ternate cultivation and medicine production and marketing,etc.In the investigation,wo found that the ecological environment and planted a greater technical are very different.In particular,germplasm sources mixed in serious condition.Finally, establishing of germplasm resources in nursery and bank.Finishing system and identification of areas of germplasm resources on Pinellia ternate.we have been collected on the 17 major Pinellia ternate germplasm for the original plant morphology,tuber shape,microscopic characteristics and the comparative study of TLC.Then,selected four different leaf types of Pinellia ternate;The cultivation tuber is grester in diameter than the wild tuber,has far exceeded the requirements Pharmacopoeia;Through different positions and different solvent extraction system launched TLC comparison,the 17 major germplasm and four different leaf types of Pinellia ternate have difference in petroleum extraction sites.Pinellia ternata as a common wild plant in China,have greater variation on shap and components,for the latter part of the evaluation of germplasm resources and good breeding to provide a reference.In germplasm evaluation studies of Pinellia ternate,the assaying methods of extractives and total alkaloid are established.The detection result indicated that extractives and total alkaloid of Pinellia tuber from Sichuan are generally higher than those from other provinces.That states the quality of Pinellia tuber produced in Si Chuan is better.It reflects the influence that trueborn medicine to the inner quality of Pinellia tuber.In Si Chuan,the producing area of SiChuanⅢand SiChuanⅤhas better Pinellia tuber,SiChuanⅢand SiChuanⅤcan as resources of the good cultivation areas.We search the fingerprint identification ways of 1H-NMR and HPLC preliminarily,their results are the same,can see the differences of different producing areas easily of the same kind of seed, illustrate Si Chuan has the steady quality,and use the HPLC fingerprint identification way to research the 4 different kinds of Pinellia ternate which are elected.We find the bamboo leaves style has less similarity to the 3 others.The results provides a scientific basis for the study Pinellia ternate germplasm resources for the comprehensive evaluation,the establishment of standards for quality and Pinellia ternate breeding research.
