Grape seed (collected from Cabernet Sauvignon) were picked from wine residues inlaboratory, and grape seed oil was extracted by ultrasonic-assisted method. This research wasdivided into three parts: Firstly, single factor of oil extraction efficiency was studied, and theconditions of grape seed oil were optimized by quadratic rotation-orthogonal combinationdesign, and quadratic regression equations were established. The optimum parameters ofextraction were found. Secondly, the physical and chemical properties of grape seed oil byultrasonic extraction were detected and compared with the traditional solvent extraction; thephysical and chemical properties of oil were also compared with national standards of grapeseed oil. Thirdly, fatty acid composition of refining grape seed oil was analyzed by gaschromatography (GC).The main results of this research are below:(1)Comparison with petroleum, acetone, ethyl acetate, n-Hexane was somewhat moreefficient in the oil extraction than others.(2) the impact of single factor was seeds-to-solvent ratio, ultrasonic power, duration timeand extraction temperature from weak to strong.(3) The optimum parameters of ultrasonic-assisted extraction was obtained: temperature,40℃; seeds-to-solvent ratio,1:9; duration time,50min; ultrasonic power,360W. Under theoptimum conditions, the extraction efficiency of grape seed oil was up to93.21%.(4) Comparison with traditional solvent extraction, the advantage of the ultrasonicextraction was relatively high extraction efficiency, time-saving and lower temperature.(5) The physical and chemical properties of refining oil were consist with the nationalstandards of grape seed oil.(6)The gas chromatography analysis showed that grape seed oil contained linoleic acid(73.70%), oleic acid (13.93%), palmitic acid (7.97%) and stearic acid (4.40%).
