In order to develop traditional Chinese medicine film-forming agent(FFA) for prevention and treatmentmastitis in dairy cattle, the optimal range of each component about FFA was acquired by single factorexperiment; with L9(3)3orthogonal test method, the optimal composing prescriptions and processingconditions were determined; and then by adhesion tests, air permeability tests and drug release tests, propertyof FFA was detected; quality control of FFA was researched by using thin layer chromatography andhigh-performance liquid chromatography, safety of FFA was also researched by using skin test and mucousmembrane test; finally, effect of FFA on dairy cattle mastitis was researched by clinic trial.The research showed the optimal volume ratio of FFA was PVA5%, Tween-801.4%-1.6%, glycerin6%~8%, carboxymethylcellulose sodium1.9%~2.1%; volume fraction of the optimal composingprescriptions was PVA: tween80: glycerin: carboxymethylcellulose sodium=5:1.5:7:2.1; the optimalprocessing conditions: Take proper amount of liquid medicine, add to5%PVA and stir, after fully swelling,put them in water bath pot,40mesh steel filter, after drop to normal temperature, add to1.5%tween80andstir, and then add to7%glycerin and stir again, after drop to normal temperature, add to4.33%butanone andstir, finally add to carboxymethylcellulose sodium with absolute ethyl alcohol dealt and stir again; adhesiontests result showed144.71g~204.68g; air permeability tests result showed7.92g/h m~2~8.46g/h m~2; drugrelease tests result showed drug release curve was y=1.0753Ln(x)+0.0534,R~2=0.9957,achieve balance ofdrug release at330min; the color of FFA was brown, texture was uniform, and PH was about6.34; thecontent of caffeic acid was less than15ug/ml, and microorganism control of FFA was up to Chinese codex2010standard; after centrifugation30min, there was not stratified phenomenon and demulsificationphenomenon; with thin layer chromatography, each component in FFA was up to the standard, and FFA wasstable; FFA had no stimulation to skin, but had a little stimulation to eyes; the recovery rate of FFA was43.48%for subclinical mastitis in dairy cattle, and prevention rate more than95%; there was no significantdifference on milk yield, milk protein, milk sugar, butterfat rate and non-fat solid.In sum, FFA had a good control effect for subclinical mastitis in dairy cattle, and preparing process wassimple, the quality could be controlled, at the same time, FFA used easily, and FFA was security, stability. |
