Objective:(1) To study the mRNA expression of TGFBI. KLK7 and BNIP3 in the oral verrucous carcinoma(OVC-CA), paracancerous (OVC-P)and normal oral mucosa tissues(OVC-N),confirming the expression of mRNA by RT-PCR; (2) To study the expression of TGFBI in the oral verrucous carcinoma,oral squamous cell carcinoma and normal oral mucosa tissues by immunohistochemistry,investigating the possible mechanism of action for TGFBI in oral verrucous carcinoma and providing the profit for treatment of oral verrucous carcinoma and oral squamous cell carcinoma.Methods:(1) The RealTime PCR was used to analy the expression of mRNA of 4 cases oral verrucous carcinoma and normal oral mucosa tissues after isolating the total RNA, data was analyzed by software;(2) The immunohistochemistry technique was used to detect the expression of TGFBI in 18 OVC tissues,26 oral squamous carcinoma (OSCC) tissues and 14 nonnal oral mucosa tissues. Statistical analysis was performed by SPSS 16.0 software.Results:(1) Expression of TGFBI,KLK7 of OVC-CA was significantly higher when compared to paracancerous (OVC-P)and normal oral mucosa tissues(OVC-N) (p< 0.05). There was no significant difference of BNIP3 expression between OVC-CA and OVC-N (p>0.05)(2) The expression of TGFBI of OVC,OSCC,NM was significantly different when totally compared (p<0.05).There was no significant difference between OVC and OSCC (p>0.05),exogenic OVC and cyst-infiltrating OVC (p>0.05)Conclusions:(1) The mRNA expression which verified the exactness of the mRNA microarray of TGFBI,KLK7 in OVC-CA,OVC-P and OVC-N were significantly different when totally compared, playing an important role in OVC tumorigenesis Possibility. (2) The expression of TGFBI in OVC,OSCC is significantly higher than in NM, showing good agreement in the gene expression between intranscription level and in translation level.
