Objective:Through the effects of compound Chinese medicine ultra-fine powder(CCMUP) on Hetian fowl growth performance, slaughter performance, meat quality, anti-oxidation indices, immune indices and blood biochemical indexes to discuss CCMUP on the health care of Hetian fowls and its possible action mechanism. Through investigating the Chinese medicine total polysaccharide (CCMTP) effects on the proliferation of peripheral T lymphocytes of Hetian fowl and the secretion level of the cytokines in vitro, further to explore the immune regulation mechanism of CCMUP under the cell level. The study results aim to provide the date references for clinical application of CCMUP.Methods:In vivo experiment 630 one-day-old Hetian fowls(male) were randomly divided into 3 treatment groups (control group, compound Chinese medicine ultra-fine powder groups, antibiotic groups)with 6 replicates in each group (n=35 animals per replicate). The control group(CG) was fed with basal diet, the compound Chinese medicine ultra-fine powder groups(CCMUPG) and the antibiotic groups(AG) received the same diet supplemented with 1% compound Chinese medicine ultra-fine powder or antibiotic of livestock, respectively. The experiment, total 130 d, was divided into 4 periods:The brood stage(1~28 d), The nursery stage(29-56 d), The maturity stage (57~105d), The fattening (106~130 d). Collcetting relevant samples, serum samples and determination of relevant indicators in the 28 d, the 56 d, the 105 d and the 130 d, respectively. In vitro experiment Utilizing the method of water extraction and alcohol precipitation to prepare the CCMTP; Utilizing the method of MTT and EL1SA to determine the proliferation of peripheral T lymphocytes of Hetian fowl and its the content of cytokine secretion, respectively.Results:In vivo experiment (1) CCMUP significantly increased the weight of Hetian fowl during the 4 periods of trial (P<0.05) and significantly increased the ADG on the 28 d (P<0.01); (2) CCMUP had significantly increased both the rate of slaughter and the rate of leg muscle (P< 0.05 or 0.01). (3) CCMUP had no impact on the brightness of meat, pH, the shearing force of 24 h and the rate of water loss (P>0.05). (4) Through CCMUP had no impact on the antioxidant indices MDA and T-AOC which in the serum of Hetian fowl, it could significantly activate the T-SOD during the earlier stage and later stage(P<0.05). (5) CCMUP had no impact on the content of NO, IgA, ALB and the enzyme activity of NOS (P> 0.05), however, it had significantly increased the serum content of IgM on day28(P< 0.01), the serum content of IgG on day56 and day105(P< 0.05), and significantly increased the serum complement content of C3, the serum complement content of C3 and C4 on day28, the serum content of lysozyme on day28, and significantly increased the serum content of TP,GLB on day105. (6) CCMUP had no impact on the serum biochemical about the content of BUN, GREA, UA, TG and the enzlyme activity of ALP, ALT (P>0.05) CCMUP had significantly increased the enzlyme activity of AST on day28 and decreased the serum content of TCHO on dayios. In vitro experiment (1) The 78.13~625 μg/mL concentration range of CCMTP could ould stimulate the proliferation of peripheral. (2) The 9.77~625 μg/mL concentration range of CCMTP could stimulate the proliferation of peripheral T lymphocytes. (3) The 19.53~78.13 μg/mL concentration range of CCMTP could markedly enhanced the peripheral T lymphocytes to secrete the IL-2, IL-4 (P< 0.05). The 78.13~312.50 μg/mL concentration range of CCMTP had enhanced the secretion of IL-6 (P< 0.05). However, the CCMTP had no impacts on the secreting of IFN~γ (P>0.05).Conclusion:The research results show that, CCMUP could increased Hetian fowl growth performance, meat quality, anti-oxidation ability, immune function. The CCMTP could enhance the proliferation of peripheral T lymphocytes and increase the cytokines secretion levels.
