Three field experiments were conducted to study the regulating technology of identification and analysis of glycosidic components as well as quality of flue-cured tobacco leaves with different genotypic flue-cured tobacco,spraying different enzyme preparations on tobacco before baking,the effect of IAA and GA3 interaction spraying on tobacco in luohe,Henan,2014.The main results are as follows:1.The flue-tobacco leaves are selected from different parts of Henan.A qualitative method was developed for the identifying and analyzing 9 main glycosidic flavor precursors from flue-cured tobacco leaves grown in Henan Province by using ultra performance liquid chromatography-electrospray ionization tandem mass spectrometry(UPLC-ESI-MS/MS).The chemical structures of 9 main glycosidic flavor precursors were determined by analysising the liquid chromatography and mass spectrogram.They are ionol-?-D-glucopyranoside and its structural isomer,chlorogenic acid and its structural isomer,neochlorogenic acid and its structural isomer,cryptochlorogenic acid,rutin,kaempferol-3-rutin,and their content were determined.This provides a theoretical basis for studying glycosidic substances and improving aroma quality of tobacco leaves.2.Different genotypes of flue-cured tobacco(six kinds of zhonyan100,yuyan10,K326,NC71,Zhongyan201,Yuyan6)were used as materials to compare and analyse the contents of physical characteristics,chemical components and glycosidic components.The results revealed that main physical characteristics,chemical components have significant differences among different genotype.And the kinds of glycosidic components in various tobacco leaves were basically the same,but the contents were fairly different.The physical characteristics and chemical components of zhongyan100,Yuyan 6 were well coordinated,and glycosidic components content of zhongyan100,Yuyan 6 were higher than others.The chemical components and glycosidic components of different genotypes of flue-cured tobacco were tested by correlation analysis,and the results revealed that nicotine,total sugar,total nitrogen,kalium chlorine showed significant positive correlation with ionol-?-Dglucopyranoside.The reducing sugar,kalium showed significant positive correlation with the structural isomer of ionol-?-D-glucopyranoside.The nicotine,total sugar,total nitrogen showed significant positive correlation with the structural isomer of neochlorogenic acid.The total sugar,total nitrogen showed significant positive correlation with rutin.At last,the glycosidic components content could be increased by improving chemical composition balance.3.The experiment was developed by spraying different enzyme preparation on the upper leaves of flue-cured tobacco before baking to study the influence of different enzyme preparation on chemical composition,free neutral and glycosidic aroma components,and it made the zhongyan100 as the material.It had five treatments,(1)distilled water(2)neutral protean with enzymatic activity 60000 U/g(3)?-amylase with enzymatic activity 3700 U/g(4)saccharifying enzyme with enzymatic activity 10 000 U/g(5)neutral protease+?-amylase +saccharifying enzyme.The results showed that different enzyme preparation could reduce the content of protein degradation in different degrees,and the mixed enzyme preparation spraying played a best role,the second is neutral protease.The different enzyme preparation can coordinate the chemical compositions of flue-tobacco leaves and increase the content of nicotine,total sugar,reducing sugar,kalium.Thus,enzyme preparation had a significant effect on protein degradation,total sugar,reducing sugar and total nitrogen of flue-cured tobacco,but its effect on potassium and chlorine of flue-cured tobacco was not significant.Also,the mixed enzyme preparation can increase the content of free neutral and glycosidic aroma components.Above all,the mixed enzyme preparation spraying played a better role than enzyme preparation spraying lonely.This provides a theoretical basis for improving the availability of upper leaves.4.The experiment was developed by spraying IAA and GA3 on the leaves of flue-cured tobacco after topping to study the influence of the interaction of IAA and GA3 on physical properties,chemical composition,free neutral and glycosidic aroma components.It had nine orthogonal treatments with three levels of IAA and GA3.Results showed that amendment of IAA to fields the same in GA3 level or application of GA3 to fields the same in IAA level improved the physical characteristics,coordinate the chemical components,increased the content of free and glycosidic aroma components of the flue-cured leaves,and the effect of higher concentration is better.The interaction between IAA and GA3 had some significantly positive effects on leaf length,leaf width,quality weight,filling value with IAA and GA3 spring single.Different level of IAA and GA3 had some very significantly effects on nicotine,total sugar,sugar-nicotine,and had some significantly on reducing sugar,kalium.Different level IAA and GA3 interaction can increase the contend of free-aroma components except for cembranoids,maillard reaction products,and the influence is significantly.The interaction of IAA and GA3 had some very significantly effects on the contend of rionol-?-D-glucopyranoside,structural isomer of neochlorogenic acid,chlorogenic acid and rutin and some significantly on neochlorogenic acid and kaempferol-3-rutin.The application of a high rate of IAA and GA3 was more conducive to improve the content of free and glycosidic aroma components than application of a low rate of IAA plus a low rate of GA3.And the affection which promoted the growth of tobacco plant and improved the quality of leaves is best of the treatment 20 mg/L IAA+20 mg/L GA3.The results provides theoretical basis for the growth and regulation of high quality flue-cured tobacco.
