OBJECTIVE:This topic through observing Si ni decoction on human lung adenocarcinoma cancer cells growth inhibition and apoptosis of A549 and influence of MAPK signal pathway,to provide experimental basis for perfecting this method anti-cancer theory,to provide scientific theory basis for further improving clinical curative effect.For giving full play to the advantages of characteristics of traditional Chinese medicine in the treatment of this disease and has the positive significance.To guide this reasonable application of Chinese medicine in clinical prevention and treatment of lung cancer.At the same time it can provide a new direction for the further research of new drugs.It is combined Chinese and westem medicine against lung cancer research a new train of thought.METHODS:Observing the early apoptosis by Si ni decoction on human lung adenocarcinoma A549 cancer cells after 24,48,72 hours by MTT.Using flow cytometry(FCM)to detect cell apoptosis rate to analysis of different doses of four decoction on human lung adenocarcinoma A549 cancer cells apoptosis.Using western blot method to detect Caspase3,8,9,Bax,Bcl-2,for Si ni decoction induced mechanism research of lung cancer cell apoptosis.Using Western Blot detection ERK,JNK,P38 lightning,P-ERK,P-JNK,P-P38 lightning,for Si ni decoction on the research on the effects of MAPK signal pathway.RESULTS:(1)Si ni decoction on human lung adenocarcinoma can restrain the growth of A549 cells Show the dependent between time and dose.To some extent four inverse soup effect time of 24 h,48 h,72 h after inhibition rate are increased,and the higher the concentration of,the inhibition rate is higher.(2)Si ni decoction can induce human lung adenocarcinoma A549 cells apoptosis.The apoptosis rate and concentration are dose dependent.To some extent,Si ni decoction high concentration group also,the higher the inhibitory rate.(3)Si ni decoction can inhibit the Erk,can cause the abnormal MAPK pathway activation were inhibited,thus inhibiting the proliferation of tumor cells;Si ni decoction can activate bax,restrain the Bcl-2,has the effect of promoting apoptosis protein kinase activation,thus inhibiting the proliferation of tumor cells.CONCLUSIONS:Si ni decoction has certain resistance to lung adenocarcinoma.It can inhibit the growth of A549 lung adenocarcinoma cancer cells and the inhibition rate to a certain extent was positively correlated with time and dose;Si ni decoction can promote the apoptosis of lung adenocarcinoma A549 cells.The apoptosis rate and dose were positively correlated to some extent.Si ni decoction can inhibit the MAPK pathway of abnormal activation effect,thus inhibiting the proliferation of tumor cells.Si ni decoction has the effect of promoting apoptosis protein kinase activation,thus inhibiting the proliferation of tumor cells.
