Influence Of Upbear The Clear And Downbear The Turbid Capsule On The Expression Of FN And Col-? Of Adenine-induced Chronic Renal Failure Rats

Objectives: The thesis' s object to observe the effect of Upbear the clear and downbear the turbid capsule on chronic renal failure that caused by adenine irrigation stomach to SD rats of general status,renal function and other relevant chemical indexes.To investigate the mechanism and treatment of upbear the clear and downbear the turbid capsule on RIF.Arounding the "ascending-descending theory",to provide experimental basis and new ideas for chronic renal failure treatment.Methods: Random eight rats from the whole sixty-eight to be the bland group,the left were divided into five groups except the bland group.To prepare 300 mg/kg·d suspension which Mixed 3% concentration of adenine and saline to irrigation stomach to SD rats.Arounding the treatment,the model group lavaged the same volume of saline,Shenshuaining Capsule group were given 0.046gmL-1,upbear the clear and downbear the turbid capsule in the low-dose group were given 0.048 gm L-1,the middle group were given 0.096gmL-1,the higher dose group were given 0.192 gmL-1once a day.After feeding 29 days,taking the Blood and kidney tissue.To examine kidney function,HE staining to observe the kidney pathologic change,using ELISA to examine the FN and Col-? levels in serum,immunohistochemical method was used to examine the FN and Col-? expression.Results:Upbear the clear and downbear the turbid capsule improved the general status,increase weight.The four treatment groups had improved much compared with model group(P<0.01).The four treatment groups had increased weight of right kidney(P<0.01),but still belowing with the model group.To compare with the model group,the four treatment groups had improved renal function,reducd the levels of Scr and BUN.There was a significant difference(P<0.01).Upbear the clear and downbear the turbid capsule had a significant rise of the FN in serumthey had a significant decrease of the Col-? in serum.There was a significant difference(P<0.01).The four treatment groups had reducd the levels of the FN and Col-? expression in the Kidney tissues.There was a significant difference(P<0.01).There was no difference between the middle group and Shenshuaining group(P>0.05).To compare with the model group,the crystal purine metabolites deposit with reddish-brown colour were improved to different extent in the Kidney tissues which from upbear the clear and downbear the turbid capsule.There was a significant difference(P<0.01).Especially the upbear the clear and downbear the turbid capsule in middle group.Conclusions:The upbear the clear and downbear the turbid capsule could reduce the excessive deposition of ECM,and also reducd the levels of the FN and Col-? expression in the Kidney tissues.They could suppressd the development of RIF.To pretect the renal function and delay CRF progression.
