To study the long-term participation in International Marathon amateurs as the research object,the application of 1H NMR(nuclear magnetic resonance)metabonomics was combined with routine urine and blood free radical metabolism and other physiological and biochemical indexes,taking Rhodiola capsule for a month in the marathon amateurs after detection before and after quantitative load urine metabolism proteomic changes,in-depth study of the marathon amateurs overall changes in the body metabolites taking supplements of Rhodiola,the specific marker for the metabolic pathway,the mechanism of Rhodiola influence metabolism of marathon athletes,guided amateur marathon athletes scientific training and nutritional supplement provides theoretical and practical reference.At random,8 participants who participated in 5 or more international marathon races were selected as subjects.Age 41 to 45 years old,healthy,male.8 subjects were divided into four groups according to the design of the subjects in four different stages,namely,a quiet control group(C),a quantitative load group(CE),a medication quiet group(MC),and a medication quantitative load group(ME).Group C was the control group,CE group for 20 km quantitative load endurance exercise;group MC,subjects continued to take a month Rhodiola supplements after quiet;group ME was the supplement of rhodiola root,one month after the quantitative load 20 km.Both group C and group MC received morning blood and morning urine,and both CE and ME groups received blood immediately after exercise and urine after 30 minutes.The first routine urine test urine samples,then using 1H NMR(magnetic resonance imaging)the metabonomics method of experiment,based on the Mest ReNova-6.1.1 software,the data is first baseline and phase adjustment,and then the SMICA-P13.0(Umetric,Sweden)of the software of multivariate statistical analysis,using partial least squares projection method(PLS-DA)for differences metabolite analysis of eachgroup.The subjects blood samples were used to measure blood free radical indexes such as MDA,SOD,GPX,T-AOC and so on.SPSS20.0 statistical software was used to examine the urinary differential metabolite and blood free radical index by T and Pearson correlation analysis.Result:(1)the data of urine before and after the experiment the quantitative exercise analysis found that,compared with the C group,CE group,urinary protein(PRO)and urobilinogen(URO)positive rate was significantly higher(P<0.05);ME group compared with MC group,ME group,urinary protein(PRO)increased significantly(P<0.05);compared with CE group,ME group of urobilinogen(URO)decreased significantly(P<0.05).The urine metabonomics results show that compared with the C group,CE group have significant differences in the urine of 9 compounds(P<0.05),the content of alanine,leucine,acetamide,glycoprotein lipid,glutamic acid,citric acid,valine,inositol content decreased,whereas lysine content was increased.Compared with CE group,ME group significantly increased valine(p<0.05),different degrees of change other 8 different metabolites,D-and glutamine and glutamate metabolism,lysine decomposition,inositol phosphate metabolism and lysine synthesis of four metabolic pathways are closely related.Through the MetaboAnalyst and the receiver operating characteristic curve(ROC)to analyze the body's metabolic differences show that glycoprotein lipid,citric acid and valine is the quantitative load lead to potential biomarkers of metabolic regulation in the important.(2)compared with group C,SOD activity,GSH-PX activity and T-AOC activity increased in CE group,and the content of MDA decreased,there was significant difference(P<0.05).Compared with CE group,ME group increased and T-AOC activity was significantly correlated(P<0.05),the content of MDA decreased,there was significant correlation between the difference(P<0.05),increased SOD activity and GSH-PX activity,but no significant correlation,that increased the activity of antioxidant enzymes of Marathon amateurs after the quantitative load,the level of lipid peroxidationdecreased.(3)correlation analysis of urine metabolites and blood free radical differences show that there is a significant positive correlation with GPX and lipid glycoprotein SOD and MDA and there was a significant negative correlation;there was a significant negative correlation with citric acid and GPX,MDA and SOD,MDA and SOD and leucine have a significant negative correlation,significant positive correlation with the presence of T-AOC.Conclusion:(1)The changes of urine protein and urine primary index of the marathon amateur athletes before and after the quantitative load exercise and after the red days were added.The urine metabonomic results suggest that glycoprotein lipid,citric acid and valine is the quantitative load lead to potential biomarkers for important metabolism regulation,Rhodiola could be affected by these potential biomarkers of metabolic pathways involving the matter so as to improve the material and energy metabolism in the body.(2)In this study,the marathon amateurs long-term endurance exercise to improve the movement of the body's antioxidant capacity;adding Rhodiola can significantly reduce blood lipid peroxide content,increase the total antioxidant capacity,promoting the body fatigue recovery.(3)There is a certain correlation between the free radical of human blood and the metabolic indexes of urine,suggesting that the balance between oxidation and antioxidation is closely related to the metabolism of substance.
