Objective:To study the long-term effects of high-voltage burns on serum PF-4 and GM-CSF in rats and the intervention effects of xuebijing and ulinastatin.Methods:According to random number table method to 350 healthy adult male SD rats were divided into five groups:false injury,electric injury group,saline group,xuebijing group,ulinastatin group,each group of 70,according to random number table method again put them into seven phase:electrical injury after the first 15 min,electric injury after 1h,8h after electricity,electricity after 24h,48h,72h after the electric and electrical injury after 1w,10 rats each time phase.The left forelimb and right hind limb of the tree were used as the"inlet"and"outlet"of the high-voltage(>1000V)current respec-tively,and the experimental electric shock system and transformer were used to shock the rats with 2KV high-voltage current(1.92±0.24)A,so as to make the high-voltage burn model.Intraperitoneal injection of saline 2mL/kg,xuebijing 10 mL/kg,1%ulinastatin(2×10~4u/kg)2mL/kg was performed in each group immediately after electroc ulinastatin on,24h,48h,72h and 1w,respectively.5~7ml blood was collected from the rat heart according to the time and phase groups,and the serum was isolated.The contents of PF-4 and GM-CSF were detected by ELISA double-antibody sandwich method.Con-duct factorial design variance analysis on the data.Results:1.The level of PF-4 in the electrocauterization group was significantly higher than that in the sham group,with significant difference(P<0.05).The level of PF-4 in the electrocauterization group reached the highest level 24hours after electrocauterization,and then gradually decreased.Serum PF-4levels in the xuebijing group and ulinastatin group were significantly lower than those in the electroc ulinastatin on group and the saline group(P<0.05),and both of them could inhibit the occurrence of PF-4 content peak at 24h after electroc ulinastatin on.There was no significant difference in serum PF-4content between xuebijing group and ulinastatin group(P>0.05).2.Compared with the sham injury group,the serum GM-CSF content of rats in the electrocauterial injury group showed a significant increase,with a significant difference(P<0.05).The content of GM-CSF in the electrocauterial injury group reached a peak 24h after electrocauterial injury,and then gradual-ly decreased,but at a slow rate.Serum GM-CSF levels in the xuebijing group and ulinastatin group were significantly lower than those in the electrocauterization group and the saline group(P<0.05),and both of them could inhibit the peak of GM-CSF 24h after electroshock.There was a significant difference in serum GM-CSF between xuebijing group and ulinastatin group(P<0.05).Conclusion:1.The concentration of PF-4 in the serum of rats after high-voltage burns showed an increasing trend compared with that before the injury,and reached a peak 24 hours after the electric injury,and then gradually decreased,which may be related to the stress response and wound inflammation of rats 24 hours after the injury.Treatment with xuebijing and ulinastatin significantly inhibit-ed the secretion of PF-4 in serum of rats,and the content of serum PF-4 was significantly lower than that of the electroshock group,indicating that xuebi-jing and ulinastatin could reduce the activation and aggregation of platelets,improve the microcirculation of the body,and reduce the inflammatory reaction after injury.2.High-voltage burns can increase the concentration of GM-CSF in the serum of rats,which reaches a peak 24 hours after the injury and then gradually decreases,which is mainly considered to be related to the inflammatory re-sponse of rats after high-voltage burns.The application of xuebijing and uli-nastatin could inhibit the secretion of GM-CSF in serum of rats and signifi-cantly reduce the content of GM-CSF in serum,and the effect of ulinastatin was significantly better than that of xuebijing,indicating that xuebijing and ulinastatin could inhibit the damage of inflammatory mediators to the body of rats and improve the microcirculation status of the body.
