Objective:To investigate the long-term effects of high voltage electric burn on serum GPIV and TSP in rats and the intervention effects of Xuebijing and ulinastatin.Methods:350 SD rats of clean grade were prepared and divided into sham-electric group,electric injury group,saline group,Xuebijing group,uli-nastatin group according to random number table.Each group was divided in-to seven time phases:15 minutes before electric injury,1 hour,8 hours after electric injury,24 hours and 48 hours after electric injury.At 72 hours and 1week after electric injury,10 rats were injected at each time phase.The left forelimb was used as the current inlet and the right hind limb was used as the current outlet only.The motor system and transformer were opened.The rats were shocked with voltage of 2KV and current of(1.92±0.24)A.The inter-vention drugs were injected into the abdominal cavity according to the groups and phases:saline 2mL/kg,Xuebijing 10mL/kg and 1%UTI(2×10~4u/kg)2mL/kg.The serum levels of GPIV and TSP were measured by enzyme-linked immunosorbent assay(ELISA).The results were analyzed by factorial design variance analysis.Results:1.The serum GPIV levels of rats in the electro-injury group were higher than those in the pre-injury 15 minutes(P<0.05)and sham-injury group(P<0.05)at 1 hour,8 hours,24 hours,48 hours,72 hours and 1 week after the electro-injury,and reached the highest level at 48 hours after the electro-injury group.The serum GPIV of rats in Xuebijing group and Ulinastatin group was lower than that of rats in electro-injury group and saline group(P<0.05),and the inhibitory effect of ulinastatin on GPIV was better than that of Xuebijing group.2.One hour,eight hours,24 hours,48 hours,72 hours and one week after electric injury,the serum TSP level of rats in electric injury group was higher than that in 15 minutes before electric injury(P<0.05)and sham electric injury group(P<0.05).There was a peak value at 8 hours and 72 hours in the electric injury group.Both Xuebijing and ulinastatin could reduce the serum TSP level after electric injury,but there was no significant difference between them(P>0.05).Conclusion:1.High voltage electric burn can significantly increase the levels of GPIV and TSP in serum of rats.The serum GPIV level increased and showed an up-ward trend after high voltage electric burn,but began to decrease after 48hours.The specific reason and mechanism of this process need further study.The serum TSP level reached the first peak at 8 hours after injury,and in-creased again at 72 hours after injury,but the range was slightly lower than that of the first one.The reason may be the activation of inflammatory reaction in burn wound and body at 72 hours after injury.2.GPIV and TSP signaling pathways in tissues and cells of the body are activated rapidly after being stimulated by high voltage electric burn,and the related gene transcription and regulation processes are initiated and gradually enhanced,thus affecting the inflammatory response and microcirculation of the body.Previous studies are not enough to clarify the specific mechanism of this process,and need to be further explored.3.Early intervention with Xuebijing injection and UTI after high voltage electric burn can significantly reduce the levels of GPIV and TSP in serum,alleviate inflammatory reaction and improve microcirculation.The anti-Infl-ammatory and improving microcirculation effects of UTI may be better than Xuebijing in septic shock stage.However,the comparison of specific effects and the mechanism of action need to be further explored.
