Objective:Through different cerebral infarction rats including frequency scalp electroacupuncture after treatment,the model of rat ethology score,HE staining results and ischemic area of VEGF protein expression changes,including optimization to improve the treatment of cerebral infarction movement disorders including frequency and improve the mechanism of action of nerve function reconstruction after cerebral ischemia,to explore the different frequency head cupping treatment effect of motor function of cerebral infarction rats.Methods:A total of 60 adult healthy SPF male SD rats were randomly divided into 6 groups with 10 rats in each group,namely the sham operation group,the blank group,the model group,the 2Hz group,the 50 Hz group,and the100 Hz group.In the model group,2Hz group,50 Hz group and 100 Hz group,middle cerebral artery occlusion(MCAO)was blocked by thread occlusion to induce focal cerebral ischemia.The 2Hz group,50 Hz group and100 Hz group were treated with electroacupuncture,and acupoints were selected:baihui and qianshencong patients were pierced through 0.5 inch on the side,treated once a day for 20 min each time for 14 consecutive days.Sham operation group,blank group and model group were not treated with acupuncture,but were only fixed in the same time.On day 1.3.5.7.14 after treatment,kaoutzanis score was used to evaluate the behavior of rats in each group.After the end of behavioral observation,HE staining was used to observe the pathological changes of cell morphology in the ischemic area,and immunohistochemistry was used to detect the expression level of VEGF protein in the ischemic area.SPSS23.0 statistical software was used to analyze the experimental data.Results:1.Behavioral scores: At different time points,rats in the blank group and the sham group could walk,open their eyes and eat by themselves.Compared with the blank group and the sham operation group,the behavioral score of themodel group was lower,and the difference was statistically significant(P<0.01),indicating that the rats had different degrees of neurological impairment after the model construction.On the 1st day of treatment,compared with the model group,the efficacy of electroacupuncture in the 2Hz group,50 Hz group and 100 Hz group was not obvious in behavioral performance.The rats in the three groups were unable to walk,open eyes by sound stimulation or open eyes by pain stimulation,and unable to eat.On the third day of treatment,compared with the model group,the motor function scores of the 2Hz group,50 Hz group and 100 Hz group were all higher than those of the model group,suggesting that the efficacy of electroacupuncture began to be prominent in behavioral performance,and the behavior of rats in the three groups was improved to different degrees.Treatment 5 d,group 2 Hz,50 Hz and 100 Hz group compared with model group,the movement function score electric acupuncture group were higher than that of model group,2 Hz group and 50 Hz and 100 Hz group compare,2Hz group score higher than 50 Hz and 100 Hz group,compared with 50 Hz and 100 Hz group there was no statistically significant difference(P>0.05),prompted the curative effect of different frequency differences in behavior began to appear on the system performance,to walk in rats,open and eating aspects are improved,with 2 Hz group improved the most obvious.Treatment 7 d,2 Hz group,50 Hz,100 Hz group compared with model group,the movement function score electric acupuncture group were higher than that of model group,the difference is statistically significant(P<0.01)and 2 Hz group and 50 Hz and 100 Hz group compare,2 Hz group score higher than 50 Hz and 100 Hz group,difference has statistical significance(P<0.01)50 Hz and 100 Hz group than there was no statistical significance(P>0.05),the difference of 2 Hz group rats can walk freely,to open and free to eat,Rats in the 50 Hz group and the 100 Hz group had difficulty walking,open eyes and eat freely due to sound stimulation.Treatment14 d,2 Hz group,50 Hz,100 Hz group compared with model group,the movement function score electric acupuncture group were higher than that of model group,the difference was statistically significant(P<0.05),and 2 Hz group and 50 Hz and 100 Hz group compare,2 Hz group score higher than 50 Hz and 100 Hz group,difference has statistical significance(P<0.01)compared with 50 Hz and 100 Hz group there was no statistically significant difference(P>0.05),the performance of 2 Hz group rats can walk freely,to open and free to eat,Rats in the 50 Hz group and the 100 Hz group had difficulty walking,open eyes and eat freely due to sound stimulation.This indicated that electroacupuncture treatment could improve motor function defects in MCAO model rats,and the 2Hz electroacupuncture group had the best effect.2.HE staining:Groups of rats brain tissue after 14 days HE dyeing observation of ischemic neurons form,the blank group and the control group of normal rats cortex form uniform dyeing,dense structure clear,nerve cells into granule,the cone shape and irregular shape,pink cytoplasm and nucleus aizen,outline clear,neurons without distribution density of death;Very few inflammatory cells were observed in the sham group.Model group can be obviously infarcts,regional cerebral cortex of rats and survival of nerve cells around infarcts number decreased significantly compared with the blank control group differences statistically significan(P<0.01),and the neuron structure is not clear,pale cytoplasm dye,the nucleus pycnosis,loose cytoplasm,cell body to reduce the deformation nucleus dissolved,glial cell proliferation,capillary lumen obvious deformation,even collapse.2 Hz group,50 Hz and 100 Hz group electric acupuncture cupping treatment group rats the damage degree of the ischemic cortex significant reduction compared with model group,ischemic cortex brain structure disorder and interstitial edema,relieve,cortex structure is not complete but visible pycnosis neurons,ischemic area decreases,and significantly reduce the dead cells,cell degeneration canalleviate,surviving nerve cells more differences statistically significant compared with model group(P<0.01).Compared with the electroacupuncture groups of 2Hz,50 Hz and 100 Hz,the cell body shrinkage of neurons in the 2Hz group was lighter,and there were fewer necrotic cells.The number of surviving nerve cells was significantly higher than that in the 50 Hz group and the 100 Hz group(P<0.01),and the difference was statistically significant.There was no significant difference in the number of neurons in the cerebral ischemia region between the 50 Hz group and the 100 Hz group(P>0.05).3.Expression of VEGF in ischemic cerebral cortex of rats:Under normal circumstances,there is only a small amount of VEGF in the brain tissue of rats.VEGF protein expression increased significantly after cerebral infarction.There was no significant difference between the blank group and the sham group(P>0.05);Compared with the blank group and the sham operation group,VEGF in the model group was significantly increased,and the difference was statistically significant(P<0.01).Compared with the model group,VEGF in the 2Hz group,50 Hz group and 100 Hz group was higher in each electroacupuncture group than in the model group,and the difference was statistically significant(P<0.01).Compared with the 50 Hz group and the 100 Hz group,the VEGF in the 2Hz group was significantly higher than that in the 50 Hz group and the 100 Hz group,with a statistically significant difference(P<0.01).There was no statistically significant difference between the 50 Hz group and the 100 Hz group(P>0.05).Conclusion:1.Different frequency electroacupuncture could improve the motor function of rats with cerebral infarction,and the effect was significant in the2 Hz electroacupuncture group under the condition of this experiment.2.Scalp electroacupuncture can significantly reduce the ischemic area of cerebral infarction rats and increase the expression of VEGF,scalp electroacupuncture may play a role in brain function reconstruction by promoting vascular endothelial regeneration after infarction.
