Yellow pigment from Gardenia jasminoides var.radicans is the main bioactive component found in the fruit of G.jasminoides,and it is a kind of carotenoid with good solubility in water.Gardenia yellow pigment has excellent dyeing performance,good nutritional value and health care effect,and can be widely used as a colorant in food,medicine,cosmetics,and other industrial fields.But at present the market gardenia yellow pigment product color stability is low,and gardenia yellow pigment has poor heat resistance and light resistance,and is easy to fade,so that the color stability and shelf life of the pigment product are adversely affected during the preservation and processing of industrial production,which limits the use of gardenia yellow pigment.In addition,the structure of each component of gardenia yellow pigment is different,and the exploration of the relationship between the stability and structure of monomer compounds with different structures is beneficial to improve its stability.In addition,the gardenia yellow pigment each component structure is different,the exploration of the relationship between stability and structure of the monomer compound of different structure is beneficial to improve its stability.Therefore,it is important to develop the refining and stabilization process of yellow pigment from G.jasminoides var.radicans in depth and study the stability of its chemical composition.This project mainly includes two parts:one is to take the fresh fruit of natural G.jasminoides as the raw material and carry out the research on the refining process of large pore resin adsorption on it;the second is to take the refined yellow pigment of natural G.jasminoides as the raw material and carry out the research on spray drying,freeze drying process and its chemical composition.The research results mainly include the following aspects:1.The fresh fruit of G.jasminoides var.radicans as raw material,refined yellow pigment from G.jasminoides var.radicans by adsorption of macroporous resin,the experimental result shows that the best process condition of macroporous resin adsorption refining is as follows:the macroporous resin selects LSA-10,the eluent selects 80%ethanol solution when desorption,the concentration of dynamic adsorption on the sample solution is diluted five times compared to the pigment mother solution(OD value is 0.919 after dilution 100 times).The impurity gardenia glucoside in the refined yellow pigment from G.jasminoides var.radicans solution decreased 82.61%compared with that before refining,and chlorogenic acid decreased 78.16%compared with that before refining;meanwhile,the color value after refining was 534.47,which increased 13.80 times compared with 38.74 before refining,and the refining effect was good.2.With refined yellow pigment from G.jasminoides var.radicans as raw material,yellow pigment from G.jasminoides var.radicans microcapsule powder was prepared by spray drying,and the experimental results showed that the best process conditions of spray drying were as follows:the wall material was selected from maltodextrin/gum arabic composite wall material,the core wall ratio was 1:10,the wall material ratio was 1:1,the concentration of wall material was 10%,and the inlet air temperature was 160℃;the yield of spray-dried microcapsules obtained under this process condition was The yield of spray-dried microcapsules obtained under this process was 58.9%,the embedding rate of spray-dried microcapsules reached 88.31%,and the pigment residue rate of spray-dried microcapsules reached 89.4%.Compared with the pigment powder without wall encapsulation,the encapsulated pigment residual rate was improved by 18.92%,which had a good effect on pigment protection.3.Refined yellow pigment from G.jasminoides var.radicans was used as raw material to prepare yellow pigment from G.jasminoides var.radicans microcapsule powder by freeze-drying,and the experimental results showed that the best process conditions of freeze-drying were as follows:the wall material was selected from maltodextrin/gum arabic composite wall material,the core-to-wall ratio was 1:10,the wall material ratio was 1:1,and the concentration of wall material was 10%;the encapsulation rate of freeze-dried microcapsules obtained under this process condition reached 98.22%.Under the condition of continuous treatment with 60 W light for 48 h,the pigment residual rate after embedding was enhanced by 18.77%and the color difference△E was reduced by 20.18 compared to the pigment powder without embedding with wall material,which provided good protection to the pigments.4.The refined yellow pigment from G.jasminoides var.radicans was used as the material,and the silica gel column chromatography was performed with ethyl acetate-methanol system as the eluent,and the structure identification was carried out by 1H-NMR,13C-NMR and HR-ESI-MS,as well as by searching with relevant databases and literature comparison.Crocin-2,crocin-3 and all-trans-crocetinβ-D-glucosyl were isolated from refined yellow pigment from G.jasminoides var.radicans.The stability of the crude extract of the pigment extract and each monomer compound was further investigated under 60 W light conditions,and the results showed that the compounds with symmetrical structures were more stable under light conditions,and the stability of the compounds decreased with increasing polarity.The above results will provide the theoretical basis for the refinement of yellow pigment from G.jasminoides var.radicans and the production of its microencapsulated products,and lay a certain foundation for the development of yellow pigment from G.jasminoides var.radicans in functional food and drug and other fields.
