Background: Cervical squamous cell carcinomas(CSCC), highly malignant, invasive and with property of fast metastasis , is the most common malignant tumors in gynecologic clinic which seriously affect the women's health. It's incidence tends to increase with the patients'ages dropping in recent years. Although the therapy of this tumor is improved markedly, the patients have a still high recurrence and even a bad prognosis, which is believed to owe to the invasion and metastasis of the tumor. The metastasis and invasion of the cervical squamous cell cancer have disturbed surgery for a long time. For their properties of invasion, the therapeutic efficacy of traditional operation, chemotherapy and radiotherapy is not satisfactory .Therefore, the early-stage therapy directed at it's ivasion and metastsis is what really counts for the cervical carcinoma heal. In recent years board researches about the mechanism of tumors'invasion and metastasis have been conducted, and the latest research findings suggests that the extracellμlar matric (ECM),a specific substrate for matrix metalloproteinases (MMPs), plays an key role in the invasion and metastasis. MMP-9,an important member of MMPs , can dissolve collagen IV which is the main ingredient of extracellμlar matrix and basement membrane, and make the tumor cells break their original sites to bring about the invasion and metastasis. CD147 is a member of the immunoglobμlin superfamily and is a highly glycosylated cell surface protein. CD147 is an molecμler which wildly express on the surface of many tumors, and has been confirmed by recent findings to be able to in multiple ways stimμlates fibroblasts which sorund the tumor to secrete matrix metalloproteinases (MMPs) for a easy invasion and metastasis of malignant tumors. The capacity of invasion and metastasis is closely related to the capacity of CD147 to induce MMPs. In addition ,it's proved by researches that MMP9 and CD147 can through many pathes regμlate the segmentation,differentiation ,apoptosis and adhesion of cancer cells,which thereby indicates that MMP9 and CD147 may participate the course of bad biologic behavior of cervical SCC. The profound molecular mechanism study on the bad biologic behavior ,especially the invasion and metastasis, of cervical SCC is of theoretical and clinical pratical value in supplying atheory basis for making out reasonable and effective treating scheme. This study explored the function and interaction of MMP-9 and CD147 in the biological behavior of cervical squamous cancer.Objective: To investigate the correlation between the expression of MMP9,CD147 and the metastasis, clinic stages and differentiation degree of CSCC by detecting respectively the expression of MMP9 and CD147 in cervical squmou cell carcinoma tissues and that in control group,Methods : The expression of MMP9mRNA in cervical squmous cell carcinoma tissues was detected with RT-PCR ,and the expression of CD147 in cervical squmous cell carcinoma tissues was detected with immunohistochemistry named MaxvisonTM method.Results: 1. The relative level of expression of MMP9mRNA in tissues of cervical squmous cell carcinoma and control group was respectively 0.9336±0.2343 and 0.3759±0.0882. The expression of MMP9mRNA in cervical squmous cell carcinoma tissues was noticebaley higher than that in control group and the difference had statistic significance(P<0.05); 2. The mean density of expression of CD147 in tissues of cervical squmous cell carcinoma and control group was respectively 0.2531±0.0761 and 0.1162±0.0326. The expression of CD147 in cervical squmous cell carcinoma tissues was noticebaley higher than that in control group and the difference had statistic significance(P<0.05); 3. The relative level of expression of MMP9mRNA in cervical squmous cell carcinoma tissues with lymph nodes metastasis and those without lymph nodes metastasis was respectively 1.0533±0.2417 and 0.8356±0.1797 .The expression of MMP9mRNA in cervical squmous cell carcinoma tissues was clearly higher than that in those without lymph nodes metastasis,and the difference had statistic significance(P<0.05); 4.The mean density of expression of CD147 in cervical squmous cell carcinoma tissues with lymph nodes metastasis and those without lymph nodes metastasis was 0.3051±0.0671and 0.2104±0.0536 respectively.The expression of CD147 in cedrvical squmous cell carcinoma tissues with lymph nodes meatstastis was obviously higher than that in those without lymph nodes meatstastis and the difference was significant; 5.There was no distinct difference of the expression of MMP9mRNA and CD147 between stageⅠCSCC and stageⅡCSCC; 6. The expression of MMP9mRNA was also higher in CSCC tissues of low differentiation degree than that in CSCC tissues of media differentiation degree(0.9932±0.2310 and 0.8343±0.2110 respectively) ,and the difference had statistic significance,but the difference of the expression of CD147 in these two groups had no significance;7.The expression of CD147 was positively related to that of MMP9mRNA in cervical carcinoma tissues.Conclusions: 1. The expression of MMP9 and CD147 in cervical CSCC tissues was generally high, which indicates that MMP9 and CD147 involves the occurrence of cervical squemous cell carcinoma; 2. The expression of MMP9 and CD147 was posi- tively related to the lymph nodes metastasis of CSCC; 3.There is no definite correlation between clinical FIGO stage of CSCC and the expression of MMP9 and CD147 ,however,much has to be done to analyse the unclear relationship between the differentiation degree of CSCC and the expression of MMP9 and CD147;4.The expression of CD147 was positively related the expression of MMP9 in CSCC tissues,which helps to judge the early metastasis of cervical squmous cell carcinoma and to instruct the effective clinic therapy for it.
