The Mechanical Study On The Intervention Of Combination With Toxin-resolving And Blood-activating Herbs In The Atherosclerosis Of Stagnation-poison Model From Apolipoprotein E Gene Knockout Mice

Objective:Compared with Xuezhikang,to observe the effect of different doses of pieplant and red rice that were combination with toxin-resolving and blood-activating herbs on blood lipids, aorta atherosclerosis,blood viscosity,inflammatory factors and apoptosis of stagnation-poison model from apolipoprotein E gene knockout mice.Methods:Six–week old ApoE(-/-) mice were divided randomly into six groups:stagnation-poison model group,high-dosage-treatment group(combination of pieplant and red rice),middle- dosage-treatment-group(combination of pieplant and red rice),low-dosage-treatment- group (combination of pieplant and red rice), model-comparison group, Xuezhikang group. Six-week-old heathy mice (C57BL/6J mice) were the normal control group.The mice of the first 5 groups were injected with endotoxin through enterocoelia.At week 8, the serum lipid TC,TG,LDL-C,HDL-C,blood viscosity,TNF-α,IL-1and IL-6 were measured,and the aortas and the hearts were preceded in the formalin for photograph analysis. Finally,the apoptosis regulatory protein Bcl-2 and Bax were assayed through immunohistochemical method. Results: pieplant and red rice can decrease blood viscosity, TC,TG,LDL-C,TNF-α,IL-1and IL-6,increase HDL-C, weaken bcl-2, enhance bax, protect the structure of aorta and cardiac muscle, antagonize the formation of the athrosclerotic plaque of stagnation -poison model from apolipoprotein E gene knockout mice. Conclusions : Our research indicated that combination of toxin- resolving and blood-activating herbs (pieplant and red rice)had an exact effect of reducing blood lipid, anti-inflammation, reduction of viscosity of blood, restraining apoptosis,protecting the structure of aorta and cardiac muscle, etal.It had great value in treating and preventing atherosclerosis,ACS, etal.
