Objective: To observe the changes of TNF-α,Bax,Bcl-2 mRNA and protein expression in the myocardium of rats during ARDS by oleic acid and observe the change of TNF-αprotein expression after intervention with 654-2. Methods: Eighty healthy Wistar rats were randomly divided into control group,OA2h,OA4h,OA6h group and intervention group with anisodamine(OA+654-2 group), The oleic acid was adopted to build ARDS model in OA groups from caudal vein, the OA+654-2 group was injected anisodamine (654-2) from the sublingual vein ten minutes before injecting oleic acid,control group was injected normal sodium from caudal vein.±LVdp/ dtmax,LVSP were observed at different time points and blood gas was detected,the W/D of lung was measured. TNF-α,Bax,Bcl-2 mRNA expression in the myocardium of OA and control group was detected by RT-PCR, TNF-αprotein expression in the myocardium of each group was detected by Westen blot. Results: Compared with control group, PaO2 of each OA group decreased; W/D of each OA group increased.±LVdp/ dtmax and LVSP of each OA group decreased, LVSP and±LVdp/ dtmax of OA+654-2 group were higher than that of each OA group, With time passing; TNF-αmRNA expression in myocardium gradually increased compared with control group, the change of BAX/Bcl-2 mRNA in each OA group was higher than that of control group, TNF-αprotein expression in the myocardium of OA4h,OA6h group was higher than that of control group and decreased in OA2h group. TNF-αprotein expression in the myocardium of OA+654-2 group was less than that of each OA group. Conclusions: Functional disturbance of heart may take place during experimental ARDS by oleic acid, with time passing, the expression of TNF-αmRNA and protein were increased,Bax/ Bcl-2 mRNA expression was progressly increased ,TNF-α,Bax,Bcl-2 may play a role in the functional disturbance of heart during experimental ARDS by oleic acid, 654-2 may influence the TNF-αexpression in the myocardium of rat and improved the function of the left heart during experimental ARDS by oleic acid.
