Objective: To detect the expressions of Runx3(runt-related transcription factor 3 gene), CerbB-2(Her-2) and P-gp(P-glycoprotein) in human breast carcinoma; to investigate the correlation between their expressions and the age of paients, the neoplasm tissue diameter, differentiation , lymph node metastasis , its clinical stage.Methods: HE staining and streptavidin peroxides immunohistochemistry staining were used to detect the expressions of Runx3, CerbB-2 and P-gp in the samples from 80 cases of breast carcinoma and 55 cases of surgical marginal tissue. Also , statistical analysis was used.Results: 1. 45 of 80 samples from the breast carcinoma were detected positive staining of Runx3 (56.25%), while 45 of 55 samples from surgical marginal tissue (81.82%)were detected positive staining. What's more, the decreased expression of Runx3 was significantly correlated with the neoplasm tissue diameter, differentiation , lymph node metastasis and its clinical stage in breast carcinoma(P <0.01),but not with the age(P >0.05).2. 30 of 80 samples from the breast carcinoma (37.50%) were detected positive staining of CerbB-2, while 11 of 55 samples from surgical marginal breast tissue (20.00%) showed positive staining of CerbB-2. And the overexpression of CerbB-2 was markedly correlated with the neoplasm tissue diameter, differentiation, lymph node metastasis and its clinical stage (P <0.05), but not with the age (P >0.05).3. 29 of 80 samples from the breast carcinoma (36.25%) were detected positive staining of P-gp, while10 of 55 samples from surgical marginal breast tissue (18.18%) showed positive staining of P-gp. And the expression of P-gp was markedly correlated with differentiation, lymph node metastasis and its clinical stage (P <0.05), but not with the age and the neoplasm tissue diameter. (P >0.05).4.Also,the expression of Runx3 was reversely correlated with the expression of CerbB-2 and P-gp respectively(r=-0.243, P =0.005; r=-0.219,P =0.011).And the expression of CerbB-2 was positively correlated with the expression of P-gp.(r=0.203,P =0.018).Conclusion: 1.Decreased expression of Runx3 was found in breast cancer and significantly correlated with the neoplasm tissue diameter, differentiation, lymph node metastasis and its clinical stage.2.The expression of CerbB-2 was markedly up-regulated in breast cancer, and the overexpression was correlated with the neoplasm tissue diameter, differentiation, lymph node metastasis and its clinical stage.3.The expression of P-gp was markedly up-regulated in breast cancer, and the verexpression was correlated with differentiation, lymph node metastasis and its clinical stage.4.The expression of Runx3 was reversely correlated with the expression of CerbB-2 and P-gp respectively in breast cancer. Jointly detection of Runx3, CerbB-2 and P-gp may be helpful to evaluate the biological behavior and predict the prognosis of breast cancer.
