In2010, edible fungi planting bags of Pleurotus geesteranus in Luoyuan county of Fuzhou city have appeared extensive green mold pollution,mainly of the bags were produced by Hua Yuan company.Take the Hua Yuan bags as testing material, use the bags produced by Xin Yong in the same period and the Pl.gl5persevered in the laboratory as comparison strains.This experiment has measured the moisture contents pH value and other data in the bags, studied on the Pleurotus geesteranus strains separated and purified from the bags in cultivar and fruiting experiments, and carried out the poisonous test on the Pleurotus geesteranus strains by the dsRNA technology.Discuss the cause of this serious diseases phenomena according to all the experimental results. Finally, through the research on the degeneration phenomenon of Hua Yuan strains, Choose3strains with poisonous to detoxicate and rejuvenate by the method of picking up mycelia、substrate mycelia and aerial mycelium。The main research results are as follows:1The result is that the moisture content of the edible fungi planting bags produced by Hua Yuan Company is comparatively high, between68.73%-72.93%while that by Xin Yong Company is62.33%-68.90%. The pH value of Hua Yuan bags is4.82-5.47, which is lower than that of Xin Yong bags with5.35-5.80. All the edible fungi planting bags collected have been detected bacterial contamination but free of mould.2By testing the Pl.g5and Pl.g6separated and predicated from Xin Yong bags, the Pl.g9、Pl.g13and Pl.g20from Hua Yuan bags, and the Pl.g15persevered in the laboratory in dsRNA technology, we have found all of them had virus bands with different sizes and numbers. As for the above six strains, we have analyzed the polymorphism with39primers by using RAPD、ISSR and SRAP molecular markers,found that there was no specific bands between them, from which we can say the six strains have close genetic relationships.The experiment has also carried out the other tests of the above six strains, including the colonial morphology comparison, the optimum growth temperature screening, the optimal growth pH value screening, the antagonistic reaction, the fruiting experiments and etc. cultivar experiments.The result is that there are some differences between them. Although the six strains have the same source with original name of Luoyuan P1.g57, but some of them have already mutated.3Use the dsRNA technology to detect the poison on the eight detoxificated and rejuvenated strains, we have discovered that by picking tip mycelia can reach the effect of escaping poison, while by picking substrate mycelia and aerial mycelium can’t do. Then take biological characteristics research and fruiting experiment on the8the eight detoxificated and rejuvenated strains and3starting strains.The result indicated that all the strains have the slight difference in the colonial morphology,and picking tip mycelia and substrate mycelia can obviously enhance the growth speed and the biomass of the mycelia, and shorten the growth cycle and increase production while the effect by picking aerial mycelium is slightly less than the original strain in growth speed and other aspects.By concluding all the experimental results to discuss the cause of this serious diseases phenomena, the essay believes that it has a direct connection with the strain quality, higher water content, low pH value in the edible fungi planting bags and other internal factors, and have an indirect connection with high temperature and etc. external factors. Through the purification and regeneration research, the results showed that picking tip mycelia can make the strains to escape poisonous and rejuvenate, and continuous picking substrate mycelia can also achieve regeneration effect while continuous picking mycelia may degenerate the strains.
