The phenomenon of black mushroom, mycelium atrophy, fruiting body deformity,etc.in lentinus edodes was appeared in Fujian Province in recent years,causing enormous losses.Researchers think that this phenomenon may relate to a lentinus edodes virus.Other researchers think that it also may relate to the pseudo-gypsum、the pseudo-bran and the tremendously changeable temperature of the mushroom house.The research isolated pure the lentinus edodes strains from the contamination bags in Shanghang County.We found that the strains carried with the dsRNA virus by detecting.For the discussion of the reason of the lentinus edodes contamination bags,The experiment was designed according to four factors and three levels orthogonal table.The results have shown that the quality of gypsum and whether strains with virus affect obviously the hyphae growth of lentinus edodes.The results of the cultivation have shown that the dsRNA virus is one reason of fruiting body deformity.The study explored two methods of lentinus edodes dsRNA virus detoxification, the method of hyphae tip detoxication and the method of protoplast regeneration seeding. The results have shown that the method of hyphae tip detoxication and the method of protoplast detoxification can remove lentinus edodes dsRNA virus by the detecting techniques of dsRNA, but the effect of the method of protoplast detoxification is better and gained 14 non-poisonous lentinus edodes strains. This study also did physiological and biochemical tests on different detoxification treatment on lentinus edodes. The results have shown that with the comparison of mycelial growth rate, L0183-Y03(3.2592cm/d),NX-J04-Y06(3.2600cm/d),N-JO 1-Y08(3.1816cm/d)andN-J12-Y03(3.18 56cm/d) are separately fastest growth strain corresponding to each series, with the comparison of CMC enzyme,laccase and polyphenol oxidase,L0183-Y03(0.995,55.00,2.20), NX-J04-Y06 (0.905,50.10,2.00) andN-J12-Y03(0.905,50.04,2.00) are separately highest enzyme activity strain corresponding to each series,but the significance of difference between liquid culture of biomass and xylanase is not obvious.This study also carried out molecular authentication experiment on different detoxification treatment on lentinus edodes. The results have shown that RAPD molecular marker with seven primers for analysis of polymorphism,there are no specific bands between strains with no virus and strains with virus,DNA bands remain consistency and integrity with virus-free or not in lentinus edodes.Finally, this study also carried out cultivation experiment on different detoxification treatment on lentinus edodes. In addition to fruit body morphology and contamination rate, there is little difference between the yield and the fruiting time.We screened out L0183-Y03,fuil bag time is 34d,turn color time is 26d,the weight of single flower is 17g,length of stem is 4.4cm,SHK is 1.0cm,pileus diameter is 4.8cm,pileus thickness is 1.9cm,yield per unit is 81g.Because the cultivation condition is not suitable,strains with virus trigger a viral disease, causing enormous losses.The using of strains with no virus can avoid a viral disease,it has the strong application value.
