Purpose:Rhubarb has been widely used in the treatment of CRF in early and middle stage. Many clinical and animal experiments have proved that rhubarb can protect renal function and delay the progression of CRF. The aim of this experiment is to observe different doses of rhubarb compound in the treatment of CRF's(the syndrome of deficiency of spleen and kidney qi and Turbid Toxin) clinical efficacy trends and evaluate comprehensive efficacy and safety, in order to explore the different physique more effective and less side effect of reasonable dose of rhubarb and improve clinical curative effect.Methods:According to the inclusion and exclusion criteria of this test, the 60 subjects by randomized-. controlled clinical trial methods was divided into treatment groups (low, middle, high dosage) and control group. Evaluation index of curative effect were observed before and after treatment:renal function, hemoglobin, red blood cell,24hpro,plasma albumin, Symptoms and signs of traditional Chinese Medicine. The safety index includes: liver function, electrolyte, blood coagulation function, electrocardiogram, stool and related adverse reactions.And according to the data obtained in the experiment, make preliminary evaluation of the efficacy and safety.Results:The clinical effective rate of the control group was 33.33%, The clinical effective rate was 37.50% in the low dose treatment group, the clinical effective rate was 43.75% in the middle dose group and 46.15% in the high-dose group, and there was no significant difference in the curative effect between the four groups. Because the sample size of the subjects was small, there was no significant difference in the curative effect among the groups, showing the trend of dose effect relationship. The control group and low, middle and high dose treatment groups can improve the symptoms and signs of TCM of C RF (the syndrome of deficiency of spleen and kidney qi and Turbid Toxin). In the course of treatment, there were some adverse reactions with different degrees of diarrhea, which was positively correlated with the dosage of rhubarb. Except the 2 high dose group subjects were unable to tolerate the diarrhea due to their own requirements to withdraw from the test,the subjects can be tolerated mainly by rhubarb induced diarrhea, Therefore, the clinical safety of Rhubarb Chinese herbal compound is better.Conclusion:1. Rhubarb groups of different doses have a good clinical effect on the CRF (the syndrome of deficiency of spleen and kidney qi and Turbid Toxin), which can improve the symptoms and signs of TCM. With the change of rhubarb dose, the clinical efficacy index showed the trend of dosage effective relationship. Because the sample was small, the difference was not statistically significant.2. Rhubarb groups of different doses have different levels of serum creatinine, but because of the small sample size, there was no statistical significance.3. According to the trend of comprehensive clinical curative effect and the side effect, Clinical recommended dosage of the rhubarb which was effective and has the minor side effect was 10g.
