Objective:To investigate the therapeutic effect and mechanism of Xiao Shuang decoction on treating traumatic limb deep venous thrombosis by discussing the changes of NF-KB in vein wall of rabbits with deep venous thrombosis and the effect of Xiaoshuan decoction-intervention on NF-KB and its relationship with deep venous thrombosis.Methods: 45 New Zealand rabbits were divided into five groups,randomly selected 27 wit h femoral vein clamp method to established the animal mode of deep venous thrombosisl,an d divided into Xiaoshuan decoction group(A),low molecular weight heparin group(B),mo del group(C)after the success of the building.The remaining 18 were divided into the sha m operation group(D)and blank group(E).Group A received Xiaoshuan decoction by gastric tube.Group B was treated with low molecular weight heparin by subcutaneous injection an d Group C?D?E with physiological saline by gastric tube.Detection the NF-KB p65 mR NA and IKBamRNA levels in vein wall of rabbits at 1?3?7days after modeling by RT-PC R method,then statistical analysis of experimental data.Results:(1)The levels of NF-KB p65 mRNA in Xiaoshuan decoction group,low molecular weight heparin group and model group are keeping the same trend of rising first and then down in,and the highest at 3rd day.Instead IKBamRNA keeps the opposite trend and the lo werst at 3rd day.(2)Compared with blank group,the difference of NF-KB p65 mRNA and IK BamRNA in the sham operation group was no statistically significant(P>0.05);At the same time after modeling,the levels of NF-KB p65 mRNA in XSD group,LMWH group and mo del group than those in the sham operation group was significantly increased,then IKBamR NA was decreasd,the difference was all statistically significant(P< 0.01);the levels of NFKB p65 mRNA in XSD group,LMWH group than those in the model group was significant ly decreasd,then IKBamRNA was increased,the difference was all statistically significant(P<0.01);The difference between in XSD group and LMWH group was no statistically signi ficant(P>0.05).Conclusion: The XSD can effectively reduce the inflammatory injury of bloodvessel walls by regulate NF-KB signaling pathways after the thrombus formation in the blood.Suggesting that regulation of NF-KB signaling pathway is one of the mechanisms of Xiaoshuan deco ction therapy for deep vein thrombosis.
