Pistacia chinensis Bunge is one of the important biomass energy tree species in China.And it is also an important tree species for biodiesel production.Pistacia chinensis Bunge is dioecious.But The project team found a series of Pistacia chinensis Bunge monoecious resource in their investigation about Pistacia chinensis Bunge in 2012-2013.In order to further explore the monoecious Pistacia chinensis Bunge resources,we carry out the research on the pollination technology of monoecious Pistacia chinensis Bunge.The pollen germination rate was measured in the first to last flowering stages of the monoecious Pistacia chinensis Bunge,which to determine the appropriate time for the pollon collection of the monoecious Pistacia chinensis Bunge.By comparing the germination rate of pollen at different germination temperatures in order to determine the suitable culture temperature of the monoecious Pistacia chinensis Bunge pollen.The suitable pollen storage temperature and time of the monoecious Pistacia chinensis Bunge were found under different temperature storage conditions.The different genotype of monoecious Pistacia chinensis Bunge pollen were compared to select pollination tree.The germination rates of 4 types of pollen of staminate and pistillate inflorescences borne on different brunches of the same tree,staminate and pistillate inflorescences borne on the same brunch,staminate and pistillate flowers borne in the same inflorescence and hermaphrodite were compared,the control was the male Pistacia chinensis Bunge.The father of offspring of the monoecious Pistacia chinensis Bunge was identified by SSR molecular marker technique,and count the proportion of the male parent for statistics in the offspring,so as to select appropriate pollination combinations of the monoecious Pistacia chinensis Bunge.then a new method of identifying suitable pollination combinations by DNA paternity testing is proposed.The main results are as follows:The germination rate of the monoecious Pistacia chinensis Bunge pollen collected at the flowering stage was higher than that of 25.4%and 38.6%at the first and the last flowering stage,which was 52.3%,which was significantly higher than the pollen collected in the early flowering period(P<0.05),and the suitable time for pollen collection was the flowering stage.Under the germination temperatures of 20?,25? and 30?,the germination rate of the monoecious Pistacia chinensis Bunge pollen was the highest at 30?,46.7%,and was significantly higher than that of two germination temperatures of 32.5%and 33%(P ?0.05),and the suitable germination temperature of pollen was 30?.During the 35 day storage of the monoecious Pistacia chinensis Bunge pollen,the germination rate of pollen decreased from 42.1%to 40.4%at-20?,and decreased by 1.7%.The germination rate of pollen stored under 25? decreased from 39.8%to 22.7%,and decreased by 17.1%.The germination rate of pollen stored at-20?decreased less,so the monoecious Pistacia chinensis Bunge pollen were more suitable for storage at-20 ?,and could be stored for more than 30 days.Among four different types of monoecious Pistacia chinensis Bunge pollen,the pollen germination rate of the staminate and pistillate flowers borne in the same inflorescence was 35%,which was significantly higher than that of the staminate and pistillate inflorescences borne on the same brunch,but the difference of the pollen germination rate between the two and other types of inflorescence and control was not significant.The pollen germination rates of the monoecious Pistacia chinensis Bunge F1 and T6 were 60%,62.2%,respectively,which were significantly higher than Lb3,Lb8,F2,T3,T7(P?0.05),which were significantly higher than Lb3(P<0.01),37.8%.F1,T6 and other Lb2,Lb4,F3,F4 and T2 with the least significant difference can be considered as the pollination tree.In the paternity test,a total of 45 offspring whose mother were F3 were paternal identificated.The progeny of 44.4%of the results was F2/F9.The rest of the offspring with the same paternal group accounted for 33.3%,17.8%and 2.2%,respectively.Considering the pollen volume,F2 might be more suitable for F3 pollination tree.The result of cluster analysis shows that the SSR selected from the test site is not close enough due to the close relationship between the male parent.A method of identifying suitable pollination combinations by DNA paternity testing is proposed.
