Effects Of Intracellular Ca²⁺ Concentration And Mitochondrion In MCAO Rats By TNHL Acupuncture

Objective:TNHL Acupuncture Method has been clinically proved,Therapeutic effect of cerebral infarction is remarkable.But there are still some shortcomings,molecular mechanism needs further exploration.Therefore,this topic first optimizes the stimulation parameters of acupoint selection and electro-acupuncture through animal experiments,second,To observe the effect of Tongnao Huoluo acupuncture on intracellular calcium concentration and mitochondria in MCAO rats,to explore the protective mechanism of Tongnao Huoluo Acupuncture on cerebral infarctionMethods:The experiment consists of two steps,First step,to determine the best method of acupuncture through animal experiments.There were 10 groups in total:G1(model group)、G2(scalp acupoint One,2Hz)、G3(scalp acupoint One,50Hz)、G4(scalp acupoint One,100Hz)、G5(scalp acupoint Two,2Hz)、G6(scalp acupoint Two,50Hz)、G7(scalp acupoint Two,100Hz)、G8(TNHL acupuncture,2Hz)、G9(TNHL acupuncture,50Hz)、G10(TNHL acupuncture,100Hz),intervention of electro-acupuncture on MCAO rats,before and after treatment,give the rats scored with Neurobehavioral Score,determine the best group of acupoints and the frequency of needling by the score.The second step,research on molecular mechanisms,the experiment was divided into five groups:G1(Blank group)、G2(Sham operation group)、G3(MCAO group)、G4(Sham treatment group)、G5(treatment group、the best acupoints and acupuncture frequency in MCAO rats).Five days later,measurement of intracellular free Ca2++concentration and mitochondrial membrane potential in hippocampus of rats in each group.Results:The first step:G5 group compared with model control group,There are significant differences in statistics(P<0.05).The other groups were compared with the control group,There were no significant differences.The second step,Calcium Ion Concentration Detection:Comparison of blank group and MCAO group,there are significant differences in statistics(P<0.01),comparison between treatment group and MCAO group,there were significant differences(P<0.01),the treatment group was compared with the blank group,there was no significant difference(P=0.264).Mitochondrial examination:Comparison between treatment group and MCAO group,there were significant differences(P<0.01).Conclusion:Comprehensive comparative analysis,G5 group(scalp acupoint Two,2Hz)is the best effect,that is BaiHui、TaiYang,FengFu、SuaiGu hrough JiaoSun,with 2 Hz,optimized TNHL Acupuncture,then according to measuring intracellular calcium concentration、mitochondrial membrane potential,shows that TNHL acupuncture and moxibustion can promote the recovery of neurons in MCAO rats,also proved that TNHL Acupuncture treatment of cerebral infarction may have a certain effect.
