H2O2 Responsive Cascade Tumor Drug Delivery System Based On Stenhouse Salt

Abstract/Summary:
As a new photochromic molecule,Stenhouse salt has been successfully applied in the field of tumor drug delivery due to its different structure from hydrophobic to hydrophilic after irradiation.However,the external light source is always needed to destroy the micelle structure,which is a defect of Stenhouse salt in the field of drug delivery.In order to solve this practical problem,we designed a drug delivery system based on hydrogen peroxide overexpressed in tumor microenvironment,which responded to hydrogen peroxide for self luminescence and spontaneously destroyed the micelle structure.By changing the electron donor group of Stenhouse salt,we successfully designed a Stenhouse derivative with absorption wavelength around 500 nm.Based on this,we synthesized the fluorescent group BLSA with the luminescence wavelength around 550 nm,and achieved good energy matching.By wrapping DOX in the micelles,we realized the cascade drug release after the tumor site responded to H2O2.1.Synthesis work.The micellar molecule DASA-PEG and the fluorophore BLSA were synthesized,and a complete drug delivery system CLDRS was prepared.2.Perform basic characterization work.Through the spectral characterization,on the one hand,it proved the successful loading of the drug,and on the other hand,it successfully realized the chemiluminescence generated after responding to H2O2,which was absorbed by the micelles again.Through the DLS test,the experimental group’s particle size structure dropped from 13 nm to about 3nm after adding H2O2,and the success of the cascade reaction was achieved.3.Conduct experiments on biological safety and cell applications.Micellar molecule shows good biosafety to He La and L929 cells.The complete drug delivery system CLDRS has significantly higher biological toxicity to He La cells.
