The thesis mainly reported the studies on chemical constituents of Oldenlandia diffusa (Willd.) Roxb and Polygonum cuspidatum, Which were known as folk anticancer herbs. The thesis also contains the report of the progress on chemical constituents, pharmacological studies and clinical uses of Oldenlandia diffusa (Willd.) Roxb.In our studies for the search of plant deriveted DNA-cleavage agents, we find that the ethanol extracts of Oldenlandia diffusa (Willd.) Roxb and Polygonum cuspidatum have significant DNA-cleavage activities. In order to find the new DNA-cleavage agents from these two herbs, we studied chemical constituents of Oldenlandia diffusa (Willd.) Roxb and Polygonum cuspidatum.Nineteen compounds were isolated from Oldenlandia diffusa (Willd.) Roxb by various chromatography methods, including silica gel chromatography, reversed phase silica gel chromatography, sephadex LH-20 chromatography and polyamide chromatography. Eleven compounds were elucidated by various spectrum (mainly MS, 1HNMR and 13CNMR spectrum) to be kaempferol (1), kaempferol 3-O-B-D-gluco pyranoside (2), kaempferol 3-O-(6"-O-a-L-rhamnosyl)-B-D-glucopyranoside (3), quercertin 3-O-J3-D-glucopyranoside (4), quercertin 3-O-(2"-O-B-D-glucopyranosyl)-B-D-glucopyranoside (5), E-6-O-p-methoxycinnamoyl scandoside methyl ester (6), Z-6-O-p-methoxycinnamoyl scandoside methyl ester (7), asperuloside (8), asperulo sidic acid (9), ethyl glucopyranoside (10) and daucosterol (11). Compounds 1-3, 5 and 10 were isolated from this herb for the first time.Ten compounds were isolated from Polygonum cuspidatum by various chromatography methods, including silica gel chromatography, reversed phase silica gel chromatography, sephadex LH-20 chromatography and polyamide chromatography. Their structures were elucidated by various spectrum (mainly MS, 1HNMR and 13CNMR spectrum) to be as resveratrol (PC-1), peceid (PC-2), emodin(PC-3), emodin-8-O-B-D-glucopyranoside (PC-4), 2-methoxy-6-acetyl-7-methyl-juglone (PC-5), citreorosein (PC-6), 5,7-dihydroxyl (3H)-isobenzofuranone (PC-7), gallic acid (PC-8), (+)-catechin (PC-9) and daucosterol (PC-10). Compounds PC- 1 and PC-7-PC-9 showed significant DNA-cleavage activities. And under the circumstance that 20 uM Cu2+ were activator the DNA cleavage activity of compound PC- 1 have not been reported previously. The Compound PC- 7's DNA strand scission activity under any circumstance have not been reported previously.At last, the progress on chemical constituents, pharmacological studies and clinical uses of Oldenlandia diffusa (Willd.) Roxb during the last years was simply summarized.
